morphological malformations in limbs and skeletal structures induced by retinoic acid in mouse embryo (nmri)

چکیده
retinoic acid (ra) plays a key role in pattern formation along the major body axis and limbformation during vertebrate development. exposure to excessive retinoic acid (in uterus) generatescongenital malformations in limbs, craniofacial, cns, urogenital, heart, and axial skeletons. in thesestudies, seven groups of nmri pregnant mice were administered a single gavage dose of 100 mg /kgbody weight; all-trans-retinoic acid dissolved in dmso at days 9, 10, and 11 of gestation (groups 2, 4,6), dmso at days 9, 10, and 11 (groups 1, 3, 5) and left (control group) untreated. animals weresacrificed at day 18 of gestation via cesarean. half of the total numbers (n=115) of fetuses wererandomly selected for histological studies, using hematoxylin-eosin methods, and others were used tostudy the skeletal defects. it was demonstrated that embryonic exposure to ra reveals that on day 9, 37% of forelimbs and 31% of hindlimbs have some abnormalities, and on days 10 and 11, 100% showdefects on craniofacial, vertebral column, ribs and limbs.

Purpose: To determine the inhibitory effects of eugenol on cytotoxic activities of retinoic acid in the liver tissue of the NMRI mouse. Materials and Methods: In this study, pregnant mice were administered by a single oral dose of 100 mg/kg eugenol from 5th to 10th days of pregnancy and also a single oral dose of 60 mg/kg RA at the day 10 of gestation. Pregnant females were sacrificed at the d...

Most of the systematic studies used in the development of human embryo culture media have been done first on mouse embryos. The general use of NMRI outbred mice is a model for toxicology, teratology and pharmacology. NMRI mouse embryo exhibit the two-cell block in vitro. The objective of this study was to evaluate and compare the effects of four kinds of culture media on the development of zygo...
متن کاملRetinoic acid synthesis and hindbrain patterning in the mouse embryo.
Targeted disruption of the murine retinaldehyde dehydrogenase 2 (Raldh2) gene precludes embryonic retinoic acid (RA) synthesis, leading to midgestational lethality (Niederreither, K., Subbarayan, V., Dolle, P. and Chambon, P. (1999). Nature Genet. 21, 444-448). We describe here the effects of this RA deficiency on the development of the hindbrain and associated neural crest. Morphological segme...

Retinoic acid (RA) is thought to play a role in embryonic pattern formation in vertebrates. A naturally occurring gradient of endogenous RA has been demonstrated in the developing chick limb bud, while local application of RA leads to the formation of additional digits. In mammals, a well-defined spectrum of birth defects has been reported as a result of fetal exposure to excess RA. In analogy ...

To clarify the relationship between neural crest cells and various developmental eye abnormalities, pregnant mice were administered an intraperitoneal injection of 12.5 mg/kg retinoic acid (RA) suspended in corn oil on day 7 of pregnancy (RA group). Control mice received an equal volume of corn oil only (control group). The fetuses were removed by laparotomy on day 18 of gestation. The fetal mo...
