diagnostic plausibility of mtbdrplus and mtbdrsl line probe assays for rapid drug susceptibility testing of drug resistant mycobacterium tuberculosis strains in pakistan

چکیده
conclusions the mtbdrplus and mtbdrsl genotypic testing can serve as useful additional tools for dst in a high-burden country like pakistan provided it is used in combination with phenotypic testing. results the sensitivity of mtbdrplus for isoniazid and rifampicin was found to be 88.8% and 90.2%, respectively, while sensitivity of mtbdrsl for fluoroquinolones, amikacin/capreomycin, and ethambutol was found to be 72.9%, 81.8%, and 56.6%, respectively. patients and methods this was a cross-sectional study conducted at the indus hospital, karachi. lipa testing was performed on 196 smear-positive samples using bactec mgit 960 as a gold standard. background world health organization (who) recommends the use of line probe assays (lipas) for rapid drug susceptibility testing (dst). however, only a limited number of studies from pakistan have documented the performance characteristics of line probe assays in testing multi-drug resistant (mdr) strains of mycobacterium tuberculosis (mtb). objectives the objective of this work is to evaluate the diagnostic plausibility of the lipa tests mtbdrplus and mtbdrsl on mdr mtb isolates from pakistan.

Diagnostic Plausibility of MTBDRplus and MTBDRsl Line Probe Assays for Rapid Drug Susceptibility Testing of Drug Resistant Mycobacterium tuberculosis Strains in Pakistan
Background: World health organization (WHO) recommends the use of line probe assays (LiPAs) for rapid drug susceptibility testing (DST). However, only a limited number of studies from Pakistan have documented the performance characteristics of line probe assays in testing multi-drug resistant (MDR) strains of Mycobacterium tuberculosis (MTB). Objectives: The objective of this work is to evaluat...
متن کاملRapid diagnosis of Mycobacterium tuberculosis infection and drug susceptibility testing.
CONTEXT The global control of tuberculosis remains a challenge from the standpoint of diagnosis, detection of drug resistance, and treatment. This is an area of special concern to the health of women and children, particularly in regions of the world with high infant mortality rates and where women have limited access to health care. OBJECTIVE Because treatment can only be initiated when infe...
متن کاملEvaluation of MTBDRplus and MTBDRsl in Detecting Drug-Resistant Tuberculosis in a Chinese Population
Background. This study aims to evaluate GenoType MTBDRplus and GenoType MTBDRsl for their ability to detect drug-resistant tuberculosis in a Chinese population. Methods. We collected 112 Mycobacteria tuberculosis strains from Jiangsu province, China. The conventional DST and line probe assay were used to detect drug resistance to rifampicin (RFP), isoniazid (INH), ofloxacin (OFX), kanamycin (Km...
متن کاملDiagnostic Performance of the GenoType MTBDRplus and MTBDRsl Assays to Identify Tuberculosis Drug Resistance in Eastern China
BACKGROUND The WHO recently has recommended the GenoType MTBDRplus version 1.0 and MTBDRsl version 1.0 assays for widespread use in countries endemic with drug-resistant tuberculosis. Despite this, these assays have rarely been evaluated in China, where the burden of drug-resistant tuberculosis is among the highest globally. METHODS Mycobacterium tuberculosis clinical isolates were obtained b...

October 2007, Vol. 97, No. 10 SAMJ Drug-resistant tuberculosis (TB), and especially multidrugresistant (MDR) TB (i.e. resistance to isoniazid and rifampicin with or without resistance to other drugs) or extensively drug-resistant (XDR) TB (i.e. MDR resistance plus resistance to the fluoroquinolones and one of the second-line injectables – kanamycin, amikacin or capreomycin),1 has very important...
متن کاملGenotype MTBDRplus: a further step toward rapid identification of drug-resistant Mycobacterium tuberculosis.
In a previous publication (1), we evaluated the Genotype MTBDR (GT-MTBDR; Hain Lifescience, Nehren, Germany) assay as a rapid diagnostic tool for multidrug resistance (MDR) detection in Mycobacterium tuberculosis isolates and clinical specimens. The main limitation of the test was the low sensitivity for isoniazid resistance detection (70%), due to the targeting of only one (katG) of the genes ...
