The effect of growth rate on pyrazinamide activity in Mycobacterium tuberculosis - insights for early bactericidal activity?
BACKGROUND Pyrazinamide (PZA) plays an essential part in the shortened six-month tuberculosis (TB) treatment course due to its activity against slow-growing and non-replicating organisms. We tested whether PZA preferentially targets slow growing cells of Mycobacterium tuberculosis that could be representative of bacteria that remain after the initial kill with isoniazid (INH), by observing the ...
متن کاملShort-term storage does not affect the quantitative yield of Mycobacterium tuberculosis in sputum in early-bactericidal-activity studies.
Early-bactericidal-activity (EBA) studies measure the change in mycobacterial load in sputum over time to evaluate antituberculosis drugs. We investigated whether a delay in sputum processing influences the quantitative results of sputum mycobacterial culture. We identified pretreatment smear-positive sputum samples collected overnight and processed at a single laboratory. Sputum volume, time f...
متن کاملBactericidal activity of OPC-67683 against drug-tolerant Mycobacterium tuberculosis.
OBJECTIVES There is an urgent need for drugs that hasten sterilization in tuberculosis; however, we presently lack indicators of this activity to guide early drug development. We previously described a novel in vitro assay to study mycobacterial phenotypic drug tolerance, in which sterilizing activity could be assessed. OPC-67,683 is a novel imidazooxazole that accelerates sterilization in the ...
متن کاملMagnetic bead protocol for culturing Mycobacterium tuberculosis from sputum specimens.
A magnetic bead protocol and a standard centrifugation protocol yielded Mycobacterium tuberculosis in 40/50 sputum specimens in 12 ± 1 days and 11 ± 2 days, respectively (P > 0.05 by Student's t test). Manipulation took 35 ± 5 min and 45 ± 10 min, respectively (P < 0.05). The magnetic bead protocol could advantageously replace centrifugation for culturing M. tuberculosis from sputum.
متن کاملPolymerase Chain Reaction Detection of Mycobacterium Tuberculosis from Sputum
Tuberculosis has remained to be a major public health problem in Nepal. The risk of spread of infection and emergence of drug-resistant strain has created the need for a rapid, sensitive and specific diagnostic test. In addition, clinically suspicious cases that do not give positive result in conventional laboratory test need more sensitive test for diagnosis. In order to evaluate the possibili...
