Abstract. Istiqomah L, Cahyanto MN, Zuprizal. 2022. Xylanase production by Trichoderma virens MLT2J2 under solid-state fermentation using corn cob as a substrate. Biodiversitas 23: 6530-6538. is one of non-starch polysaccharide (NSP) degrading enzyme encompass in industrial application like animal feed, food, biofuel, and textile. The objective this study was to produce characterize the crude xylanase from isolate that isolated coconut husk. Molecular identification revealed identified virens. produced (108 spores/g corncob) substrate with 80% initial moisture content (SSF) at 30°C for 7 d incubation. Changes surface morphology structure fermented were monitored Scanning Electron Microscope (SEM). Statistical analysis performed One-way variance (ANOVA) followed Tukey Kramer post hoc test compare treatment means. Under parameters maximum activity 181.22 U/g-IDW, loss dry matter 11.43%, pH decreased (4.11) 5 incubation compared (5.47). between broad range 3-8 showed acidophilic mesophilic characteristics (optimum 5,0 40°C), also conserved more than 50% before 4 h after 40°C. micrographs SSF appeared intact smoother, while turned coarser SSF, suggestive disrupted porous surface. These findings reveled T. could extracellular potential disrupt cell wall cheap thereby increased reactive area enzymatic situ hydrolysis.

Xylanase Production under Solid State Fermentation by Aspergillus niger
Central composite orthogonal design was applied to quantify relations of xylanase production, loss of dry matter and change of pH with four critical variables during solid state fermentation of a mixture of wheat bran and wheat straw on which Aspergillus niger CCUG 33991 was cultivated. The studied variables included the percentage of wheat straw, temperature, moisture content, and fermentati...
متن کاملMonascus pigment production by solid-state fermentation with corn cob substrate.
Natural pigments are an important alternative to potentially harmful synthetic dyes. We investigated the feasibility of corn cob powder as a substrate for production of pigments by Monascus purpureus KACC 42430 in solid-state fermentation. A pigment yield of 25.42 OD Units/gram of dry fermented substrate was achieved with corn cob powder and optimized process parameters, including 60% (w/w) ini...
متن کاملSaccharification and hydrolytic enzyme production of alkali pre-treated wheat bran by Trichoderma virens under solid state fermentation
BACKGROUND In continuation of our previously interest in the saccharification of agriculture wastes by Bacillus megatherium in solid state fermentation (SSF), we wish to report an investigation and comparative evaluation among Trichoderma sp. for the saccharification of four alkali-pretreated agricultural residues and production of hydrolytic enzymes, carboxymethyl cellulase (CMCase), filter pa...
متن کاملProduction of Xylanase from Arthrobacter sp. MTCC 6915 Using Saw Dust As Substrate under Solid State Fermentation
Saw dust was used as substrate for xylanase production from Arthrobacter sp. MTCC 6915. The study of period of incubation, temperature, pH, carbon, and nitrogen sources for xylanase production was optimized. Xylanase production was found to be optimum at an incubation period of 96 hrs (117.0 U/mL), temperature 30°C (105.0 U/mL), and pH 9.0 (102.9 U/mL). The results showed that the xylanase prod...
متن کاملExtracellular xylanase production by Fusarium species in solid state fermentation.
Fusarium sp. has been shown to be a promising organism for enhanced production of xylanases. In the present study, xylanase production by 21 Fusarium sp. isolates (8 Fusarium culmorum, 4 Fusarium solani, 6 Fusarium verticillioides and 3 Fusarium equiseti) was evaluated under solid state fermentation (SSF). The fungal isolate Fusarium solani SYRN7 was the best xylanase producer among the tested ...
