چکیده
Abstract Optical imaging has the potential to reveal high-resolution information with minimal photodamage. The recent renaissance of super-resolution, widefield, ultrafast, and computational methods broadened its horizons even further. However, a remaining grand challenge is at depth over widefield high spatiotemporal resolution. This achievement would enable observation fast collective biological processes, particularly those underpinning neuroscience developmental biology. Multiphoton depth, combining temporal focusing single-pixel detection, an emerging avenue address this challenge. novel physics driving approach offer great for future advances. chapter articulates theories detection details specific TempoRAl Focusing microscopy single-pIXel (TRAFIX), particular focus on current practical implementation prospects.

Fast volumetric imaging with patterned illumination via digital micro-mirror device-based temporal focusing multiphoton microscopy
Temporal focusing multiphoton microscopy (TFMPM) has the advantage of area excitation in an axial confinement of only a few microns; hence, it can offer fast three-dimensional (3D) multiphoton imaging. Herein, fast volumetric imaging via a developed digital micromirror device (DMD)-based TFMPM has been realized through the synchronization of an electron multiplying charge-coupled device (EMCCD)...
متن کاملWide-field multiphoton imaging of cellular dynamics in thick tissue by temporal focusing and patterned illumination
Wide-field temporal focusing is a novel technique that provides optical sectioning for imaging without the need for beam scanning. However, illuminating over large areas greatly reduces the photon density which limits the technique applicability to small regions, precluding functional imaging of cellular networks. Here we present a strategy that combines beam shaping and temporal focusing of am...
متن کاملImaging primaries and multiples simultaneously with depth-focusing
Seismic imaging amplitudes are extracted with the imaging conditions t = 0 and h = 0, where t = 0 means that the take-off time of the upward-coming wave is zero, and h = 0, with h the half-offset between the source and receiver position, means that the downwardgoing and upward-coming waves meet together during the wavefield extrapolation. However, h = 0 makes no sense for multiples imaging. Thi...
متن کاملObjective, comparative assessment of the penetration depth of temporal-focusing microscopy for imaging various organs.
Temporal focusing is a technique for performing axially resolved widefield multiphoton microscopy with a large field of view. Despite significant advantages over conventional point-scanning multiphoton microscopy in terms of imaging speed, the need to collect the whole image simultaneously means that it is expected to achieve a lower penetration depth in common biological samples compared to po...
متن کاملNonlinear structured-illumination enhanced temporal focusing multiphoton excitation microscopy with a digital micromirror device
In this study, the light diffraction of temporal focusing multiphoton excitation microscopy (TFMPEM) and the excitation patterning of nonlinear structured-illumination microscopy (NSIM) can be simultaneously and accurately implemented via a single high-resolution digital micromirror device. The lateral and axial spatial resolutions of the TFMPEM are remarkably improved through the second-order ...
