چکیده

Purpose To characterize vitreous microparticles (MPs) in patients with traumatic proliferative vitreoretinopathy (PVR) and investigate their role PVR pathogenesis. Methods Vitreous MPs were characterized PVR, rhegmatogenous retinal detachment (RRD) complicated control subjects by flow cytometry. The presence of M2 macrophages epiretinal membranes was measured immunostaining. cytokines quantified ELISA assay. For vitro studies, isolated from THP-1 cell differentiated M1 macrophages, termed M1-MPs M2-MPs, used. effects mechanisms M2-MPs on RPE proliferation, migration, epithelial to mesenchymal transition analyzed. Results derived photoreceptors, microglia, significantly increased comparison RRD (PVR), whereas no significance identified between the two groups. present membranes, signature markedly elevated PVR. Moreover, In analyses showed that promoted proliferation migration cells via activation phosphatidylinositol 3-kinase (PI3K)/protein kinase B (AKT)/mammalian target rapamycin (mTOR) signaling pathway. However, did not induce expression fibrotic proteins, including fibronectin, ?-smooth muscle actin, N-cadherin cells. Conclusions This study demonstrated MP shedding PVR; specifically, polarized may contribute progression stimulating migration.

Vitreous modulation of migration and proliferation of retinal pigment epithelial cells in vitro.

Retinal pigment epithelial (RPE) cell migration and proliferation are believed to play a role in the pathogenesis of proliferative vitreoretinopathy (PVR). Since PVR develops in situations where vitreous contacts the RPE, we sought to determine whether human vitreous contains factors that stimulate proliferation and migration of RPE cells. We found that postmortem human vitreous stimulates migr...

Increased Vitreous Shedding of Microparticles in Proliferative Diabetic Retinopathy Stimulates Endothelial Proliferation

OBJECTIVE Diabetic retinopathy is associated with progressive retinal capillary activation and proliferation, leading to vision impairment and blindness. Microparticles are submicron membrane vesicles with biological activities, released following cell activation or apoptosis. We tested the hypothesis that proangiogenic microparticles accumulate in vitreous fluid in diabetic retinopathy. RESE...

Macrophage Foam Cell–Derived Extracellular Vesicles Promote Vascular Smooth Muscle Cell Migration and Adhesion

BACKGROUND A new mechanism for intercellular communication has recently emerged that involves intercellular transfer of extracellular vesicles (EVs). Several studies have indicated that EVs may play a potential role in cell-to-cell communication between macrophage foam cells and vascular smooth muscle cells (VSMCs) in atherosclerotic lesion. METHODS AND RESULTS This study involved the compari...

Proliferative vitreoretinopathy.

PURPOSE. The migration, proliferation, differentiation, and adhesion of cells and other cellular functions are influenced by the surrounding extracellular matrix, in normal and wound-healing conditions. The matrix metalloproteinases (MMPs) are a family of enzymes that degrade and remodel the extracellular matrix and, thus, play a central role in the wound-healing process. Proliferative vitreore...

Macrophage migration inhibitory factor levels in the vitreous of patients with proliferative diabetic retinopathy.

AIMS To assess the potential role of macrophage migration inhibitory factor (MIF) in the pathogenesis of proliferative diabetic retinopathy (PDR). METHODS MIF levels were assayed in the vitreous and paired serum samples of 73 consecutive patients with PDR (32 eyes) and macular hole or idiopathic epiretinal membrane (controls, 41 eyes). An enzyme linked immunosorbent assay technique was used t...
