Umbilical cord blood as a promising source of NK cells for immunotherapy

چکیده
Currently, a large number of studies on genetic modification cord blood NK cells (UCB-NK) are carried out at both clinical and preclinical levels. Immunotherapy based UCB-NK has great potential for antitumor therapy. However, despite having known several advantages over peripheral (PB- NK), including high concentration in low virulence rate, predominantly characterized the scientific literature as immature low-functioning cells. In this work, we studied phenotypic characteristics possibility stimulatory compensation decreased functional activity Our revealed can be poorly differentiated weakly activated with level inhibitory receptor NKG2A activating NKG2C HLA-DR, accordingly data. Two types stimuli were chosen to stimulate freshly isolated cells: 1) 100 units IL-2; 2) combinations IL-2 K-562 feeder expressing membrane-bound IL-21 (K562-mbIL21). It was shown degranulation (LAMP-1) proliferative higher than parallel cultured ex vivo PB-NK under same conditions stimulated 7 days + K562-mbIL21. Moreover, stimulation way K562-mbIL21 seemed more perspective obtain proliferatively active compared only. Since is promising improve properties cells, retroviral transduction procedure performed study transduced day 8 cultivation. study, used targeted overexpression adaptor molecule DAP12, which involved signaling cell receptors. equal experimental volume viral particles. As result, efficiency found 4-fold Thus, appear tool further research cancer immunotherapy.

Genetically modified human umbilical cord blood cells as a promising strategy for treatment of spinal cord injury
PERSPECTIVE Genetically modified human umbilical cord blood cells as a promising strategy for treatment of spinal cord injury Spinal cord injury (SCI) continues to be a pressing health and social problem. The injury leads to neuronal and glial cell death accompanied by degeneration of nerve fibers. There are currently no particularly effective treatments. SCI causes profound disability of peopl...
متن کاملNanofiber Expansion of Umbilical Cord Blood Hematopoietic Stem Cells
Background The aim of this study was the ex vivo expansion of Umbilical Cord Blood hematopoietic stem cells on biocompatible nanofiber scaffolds. Materials and Methods CD133+ hematopoietic stem cells were separated from umbilical cord blood using MidiMacs (positive selection) system by means of monocolonal antibody CD133 (microbeads) subsequently, flowcytometry method was done to asses...
متن کاملMesenchymal stem cells from umbilical cord tissue as potential therapeutics for cardiomyodegenerative diseases – a review
Heart failure is one of the leading causes of death worldwide. End stage disease often requires heart transplantation, which is hampered by donor organ shortage. Tissue engineering represents a promising alternative approach for cardiac repair. For the generation of artificial heart muscle tissue several cell types, scaffold materials and bioreactor designs are under investigation. In this revi...
متن کاملUmbilical cord blood as a source of hematopoietic stem cells: from research to clinical application.
The clonogenic capacity of human umbilical cord blood (UCB) has been evaluated in several studies which found high numbers of primitive hematopoietic progenitor cells. Recently, UCB progenitor cells were shown to possess significant advantages over bone marrow (BM) in terms of proliferative capacity and immunologic reactivity. Therefore UCB has come to be considered an attractive source of hema...
متن کاملHuman umbilical cord blood as a potential source of transplantable hematopoietic stem/progenitor cells.
The purpose of this study was to evaluate human umbilical cord blood as an alternative to bone marrow in the provision of transplantable stem/progenitor cells for hematopoietic reconstitution. Although no direct quantitative assay for human hematopoietic repopulating cells is at present available, the granulocyte-macrophage progenitor cell (CFU-GM) assay has been used with success as a valid in...
