Transmission of Similar Mcr-1 Carrying Plasmids among Different Escherichia coli Lineages Isolated from Livestock and the Farmer

چکیده
Colistin use has mostly been stopped in human medicine, due to its toxicity. However, nowadays, it still is used as a last-resort antibiotic treat hospital infections caused by multi-drug resistant Enterobacteriaceae. On the contrary, colistin veterinary medicine until recently. In this study, 210 fecal samples from pigs (n = 57), calves 152), and farmer 1) were collected farm where E. coli harboring mcr-1–mcr-3 was previously detected. Samples plated, mcr-genes presence confirmed multiplex-PCR. Hybrid sequencing which determined location of mcr-1, other resistance genes, virulence factors. Eighteen isolates (13 calves, four pigs, one farmer) contained mcr-1 associated with plasmids (IncX4, IncI2, IncHI2), except for two that yielded chromosome. Similar distributed different lineages. Transmission most likely occurred horizontal gene transfer calf origin, since highly similar (99% coverage, 99.97% identity). Moreover, 33 factors, including stx2 Shiga toxin (STEC) detected, highlighting role livestock reservoir pathotypes zoonotic potential.

Molecular characteristics of mcr-1-carrying plasmids and new mcr-1 variant recovered from polyclonal clinical Escherichia coli from Argentina and Canada
We have characterized nine mcr-1-harboring plasmids from clinical Escherichia coli isolates previously described in Argentina and Canada. Three of these plasmids carried a mcr-1-variant called here mcr-1.5. All these E. coli isolates were not clonally related and were recovered in different years and locations. However, their mcr-1-harboring plasmids showed high identity among them and to other...

The emergence of the mobilized colistin resistance gene, representing a novel mechanism for bacterial drug resistance, challenges the last resort against the severe infections by Gram-negative bacteria with multi-drug resistances. Very recently, we showed the diversity in the mcr-1-carrying plasmid reservoirs from the gut microbiota. Here, we reported that a similar but more complex scenario is...
متن کاملRemarkable Diversity of Escherichia coli Carrying mcr-1 from Hospital Sewage with the Identification of Two New mcr-1 Variants
The plasmid-borne colistin-resistant gene mcr-1 has rapidly become a worldwide public health concern. This study aims to determine the host bacterial strains, plasmids, and genetic contexts of mcr-1 in hospital sewage. A 1-ml hospital sewage sample was cultured. Colistin-resistant bacterial colonies were selected on agar plates and were subjected to whole genome sequencing and subsequent analys...
متن کاملEmergence of plasmid-mediated colistin resistance (MCR-1) among Escherichia coli isolated from South African patients.
The polymyxin antibiotic colistin is an antibiotic of last resort for the treatment of extensively drug-resistant Gram-negative bacteria, including carbapenemase-producing Enterobacteriaceae. The State of the World's Antibiotics report in 2015 highlighted South Africa (SA)'s increasing incidence of these 'superbugs' (3.2% of Klebsiella pneumoniae reported from SA were carbapenemase producers), ...
