Transgene-Free Genome Editing for Biotic and Abiotic Stress Resistance in Sugarcane: Prospects and Challenges
نویسندگان
چکیده
Sugarcane (Saccharum spp.) is one of the most valuable food and industrial crops. Its production constrained due to major biotic (fungi, bacteria, viruses insect pests) abiotic (drought, salt, cold/heat, water logging heavy metals) stresses. The ever-increasing demand for sugar biofuel rise new pest disease variants call use innovative technologies speed up sugarcane genetic improvement process. Developing cultivars through conventional breeding techniques requires much time resources. advent CRISPR/Cas genome editing technology enables creation with improved resistance/tolerance various presence cassette inside genome-edited plants hinders commercial exploitation regulatory issues. However, this limitation can be overcome by using transgene-free techniques. Transgene-free approaches, such as delivery RNPs biolistics or protoplast fusion, virus-induced (VIGE), transient expression reagents Agrobacterium-mediated transformation other are discussed. A well-established PCR-based assay advanced screening systems visual marker system Transgene killer CRISPR (TKC) rapidly identify edits. These advancements in climate-smart that combat stresses produce good yields under ever-changing conditions.

Chimeric antigen receptor engineered-T (CAR-T) cells also named as living drugs, have been recently known as a breakthrough technology and were applied as an adoptive immunotherapy against different types of cancer. They also attracted widespread interest because of the success of B-cell malignancy therapy achieved by anti-CD19 CAR-T cells. Current genetic toolbox enabled the synthesis of CARs ...
متن کاملGenome Editing in Sugarcane: Challenges Ahead
Genome editing opens new and unique opportunities for researchers to enhance crop production. Until 2013, the zinc finger nucleases (ZFNs) and transcription activator-like effector nucleases (TALENs) were the key tools used for genome editing applications. The advent of RNA-guided engineered nucleases - the type II clustered regularly interspaced short palindromic repeat (CRISPR)/Cas9 (CRISPR-a...
متن کاملAbiotic and biotic stress combinations.
Environmental stress conditions such as drought, heat, salinity, cold, or pathogen infection can have a devastating impact on plant growth and yield under field conditions. Nevertheless, the effects of these stresses on plants are typically being studied under controlled growth conditions in the laboratory. The field environment is very different from the controlled conditions used in laborator...
متن کاملImpact of Silicon in Alleviating Biotic and Abiotic Stress in Sugarcane: a Review
For many years silicon (Si) deficiency in crops was relatively unknown and this element was widely regarded as non-essential for plant growth. Ever since the discovery by D’Hotman De Villiers, in 1937, that sugarcane growing on highly weathered soils in Mauritius could be rejuvenated, by applying finely crushed siliceous basalt, silicon has emerged as an important nutrient for sugarcane. With t...
متن کاملBreeding for Abiotic Stress Resistance: Challenges and Opportunities
Recent experimentation with transgenic plants has led to increased salinity tolerance, with emphasis on the areas of ion homeostasis, osmotic regulation and antioxidant protection. A case study of the major challenges and opportunities to improve stress tolerance in plants using salinity is presented. As different abiotic stresses are inter-related (e.g. salinity and osmotic stress), our abilit...
