چکیده

ABSTRACT Background Tissue inhibitor of matrix metalloproteinase-2 (TIMP-2) and insulin-like growth factor binding protein 7 (IGFBP7) are markers tubular stress urinary [TIMP-2]*[IGFBP7] is an established biomarker for risk assessment acute kidney injury. There no studies expression profiles or localization these in human renal tissue with confirmed disease. Methods We analysed 37 biopsies patients disease 10 non-diseased control TIMP-2 IGFBP7 using immunohistochemistry. Changes glomerular morphology were evaluated by a semi-quantitative glomerulosclerosis score (GSI) interstitial changes graded the injury (TSI) periodic acid–Schiff-stained paraffin sections. Interstitial fibrosis atrophy (IF/TA) according to Banff classification. Urinary was collected at time biopsy. Results had significantly greater from compared tissue, especially compartment. Here, detected proximal distal tubules while predominantly localized collecting ducts. Renal correlated staining intensity IGFBP7: GSI weakly (r = 0.36) 0.35) TSI 0.41) 0.43). histopathological damage but not expression. Conclusion Our findings underline role TIMP-2/IGFBP7 as unspecific marker that already use early detection

The diagnostic accuracy of urinary [TIMP-2]·[IGFBP7] for acute kidney injury in adults

INTRODUCTION Early diagnosis of acute kidney injury (AKI) remains a challenge. Recently, [TIMP-2]·[IGFBP7], which is a combination of urine tissue inhibitor of metalloproteinase 2 (TIMP-2) and insulin-like growth factor (IGF) binding protein 7 (IGFBP7), has been identified as a potential biomarker of AKI. We performed this meta-analysis to assess the diagnostic accuracy of urinary [TIMP-2]·[IGF...

Clinical adjudication in acute kidney injury studies: findings from the pivotal TIMP-2*IGFBP7 biomarker study

BACKGROUND The NEPROCHECK test (Astute Medical, San Diego, CA, USA) combines urinary tissue inhibitor of metalloproteinases-2 (TIMP-2) and insulin-like growth factor binding protein 7 (IGFBP7) to identify patients at high risk for acute kidney injury (AKI). In a US Food and Drug Administration registration trial (NCT01573962), AKI was determined by a three-member clinical adjudication committee...

Urinary TIMP-2 and IGFBP7 as Early Biomarkers of Acute Kidney Injury and Renal Recovery following Cardiac Surgery

BACKGROUND Difficulties in prediction and early identification of (acute kidney injury) AKI have hindered the ability to develop preventive and therapeutic measures for this syndrome. We tested the hypothesis that a urine test measuring insulin-like growth factor-binding protein 7 (IGFBP7) and tissue inhibitor of metalloproteinases-2 (TIMP-2), both inducers of G1 cell cycle arrest, a key mechan...

Urinary Biomarkers TIMP-2 and IGFBP7 Early Predict Acute Kidney Injury after Major Surgery

OBJECTIVE To assess the ability of the urinary biomarkers IGFBP7 (insulin-like growth factor-binding protein 7) and TIMP-2 (tissue inhibitor of metalloproteinase 2) to early predict acute kidney injury (AKI) in high-risk surgical patients. INTRODUCTION Postoperative AKI is associated with an increase in short and long-term mortality. Using IGFBP7 and TIMP-2 for early detection of cellular kid...

Urinary TIMP-2 and IGFBP7 for the prediction of acute kidney injury following cardiac surgery

BACKGROUND Acute kidney injury (AKI) following cardiac surgery is common and associated with poor patient outcomes. Early risk assessment for development of AKI remains a challenge. The combination of urinary tissue inhibitor of metalloproteinase 2 (TIMP-2) and insulin-like growth factor binding protein 7 (IGFBP7) has been shown to be an excellent predictor of AKI following cardiac surgery, but...
