Abstract RSC (Remodels the Structure of Chromatin) is a conserved ATP-dependent chromatin remodeling complex that regulates many biological processes, including transcription by RNA polymerase II (Pol II). We report contributes in generating accessible nucleosomes transcribed coding sequences (CDSs). MNase ChIP-seq data revealed RSC-bound nucleosome fragments were very heterogenous (?80 bp to 180 bp) compared sharper profile displayed inputs (140 160 bp), supporting idea promotes accessibility nucleosomal DNA. Notably, binding +1 and CDSs, but not with -1 nucleosomes, strongly correlated Pol occupancies, suggesting enrichment CDSs linked transcription. also observed associates throughout similar RSC, II-protected highly heterogenous, consistent interacts remodeled CDSs. This supported observation genes harboring high-levels their most affected ablating function.
