چکیده
Abstract While intracytoplasmic sperm injection (ICSI) is an asset in human Assisted Reproduction Technologies (ART), its outcomes, terms of blastocyst, still unacceptably low ruminants. The picture typically found ICSI derived bovine and ovine embryos asymmetry between a high activation rate, marked by pronuclear development, first cleavage rate. Abnormal centriole function has been indicated as possible factor which undermines embryonic development following ICSI, especially when Freeze Dried spermatozoa (FD) are used. In order to verify the hypothesis that dysfunction might be responsible for outcomes sheep, we have investigated micro-tubular dynamics, markedly aster nucleation, fertilized sheep zygotes with frozen/thawed (FT) FD spermatozoa; In Vitro Fertilized (IVF) oocytes were used control. nucleation was assessed at different time points IVF immune-detection ?-tubulin. Pronuclear stage, syngamy embryo assessed. No difference noticed timing microtubule elongation ICSI-FT control ones, while delay recorded ICSI-FD ones. proportion 2-pronuclear stage similar (47% 53%, respectively), both much lower comparing ones (73%). Likewise, observed minority groups (28.5% vs 12.5% ICSI-FT/FD respectively) controls (50%), number blocked (71.5% 87.5% respectively). no significant differences rate ICSI-FD, (31%, 34% 44%) respectively, blastocyst compromised groups, (10% ICIS-FD 19% 33% (P

P-24: The Effect of Oocyte Activation with DTT on Development of Ovine Embryos in ICSI
Background: The objective of the current study was to determine the effect of activation of injected oocytes with dithiothreitol on development of ovine embryos derived from in vitro-matured oocytes and intracytoplasmic sperm. The injected oocytes with non-treated sperm were activated with 1) ionomycin (Io) + 6-dimethylaminopurine (6-DMAP) 2) DTT, 3) DTT+ ionomycin (I)+6-DMAP. Materials and Meth...
متن کاملP-110: In Vitro Developmental Competence of ICSI-Derived Activated Ovine Embryos
Background: This study was designed to respond to the controversy regarding the necessity of oocyte activation after the ICSI procedure for normal embryo development in sheep. Materials and Methods: The effect of chemical stimulation with either 5mM ionomycin (I) for 5 minutes or ionomycin + 2 mM 6-dimethylaminopurine (6-DMAP) for 3 hours on the efficiency of ICSI, was compared in six experimen...
متن کاملGrowth rate of human preimplantation embryos is sex dependent after ICSI but not after IVF.
BACKGROUND There is concern that IVF and/or ICSI might have an adverse effect on embryonic development via epigenetic alterations. Such alterations might also be involved in the sex-related growth differences in preimplantation embryos found in some animal species. In the present study we analysed cell numbers of human male and female surplus embryos that developed to the blastocyst stage after...

in the present study, the effect of e-64 at different concentrations (0.5, 1, 5 and 10 µm) added to (1) the ivm medium on oocyte nuclear maturation and developmental competence of ovine oocytes, and (2) to the ivc medium on embryonic development of ovine embryos were investigated.
