The Haves and Have-Nots: The Mitochondrial Permeability Transition Pore across Species

چکیده

The demonstration that F1FO (F)-ATP synthase and adenine nucleotide translocase (ANT) can form Ca2+-activated, high-conductance channels in the inner membrane of mitochondria from a variety eukaryotes led to renewed interest permeability transition (PT), increase mediated by PT pore (PTP). is Ca2+-dependent mitochondrial whose function underlying molecular mechanisms have challenged scientists for last 70 years. Although most our knowledge about PTP comes studies mammals, recent data obtained other species highlighted substantial differences could be perhaps attributed specific features F-ATP and/or ANT. Strikingly, anoxia salt-tolerant brine shrimp Artemia franciscana does not undergo spite its ability take up store Ca2+ mitochondria, anoxia-resistant Drosophila melanogaster displays low-conductance, selective Ca2+-induced release channel rather than PTP. In provides mechanism cytochrome c proapoptotic proteins mediates various forms cell death. this review, we cover (or lack thereof) yeast, melanogaster, Caenorhabditis elegans, discuss presence intrinsic pathway apoptosis We hope exercise may help elucidate function(s) possible role evolution inspire further tests define nature.

The haves and the have nots

As the equipment needed to perform state-of-the-art research in many areas of biology becomes ever more expensive, Eve Marder worries that researchers in less wealthy institutions might be left behind.

Stereo Vision: The Haves and Have-Nots

Animals with front facing eyes benefit from a substantial overlap in the visual fields of each eye, and devote specialized brain processes to using the horizontal spatial disparities produced as a result of viewing the same object with two laterally placed eyes, to derived depth or 3-D stereo information. This provides the advantage to break the camouflage of objects in front of similarly textu...

The mitochondrial permeability transition pore.

This chapter reviews recent advances in the identification of the structural elements of the permeability transition pore. The discovery that cyclosporin A (CsA) inhibits the pore proved instrumental. Various approaches indicate that CsA blocks the pore by binding to cyclophilin (CyP)-D. In particular, covalent labelling of CyP-D in situ by a photoactive CsA derivative has shown that pore ligan...

Mitochondrial permeability transition pore and postconditioning.

Postconditioning has recently been described as a powerful cardioprotection that prevents lethal reperfusion injury. Growing evidence suggests that mitochondrial permeability transition may be a key event in postconditioning. This proposition arises from the complementary observations that: (1) conditions for the mitochondrial permeability transition pore (mPTP) opening are built up during earl...

Mitochondrial permeability transition pore: an enigmatic gatekeeper

Mitochondrial permeability transition pore (MPTP) is a transient structure formed in the inner mitochondrial membrane (IMM) upon oxidative challenge of the organelle. The transition is a Ca 2+ -dependent increase of permeability of IMM leading to loss of transmembrane potential (∆ψm), mitochondrial swelling and, finally, rupture of outer mitochondrial membrane leading to release of factors caus...
