The Electrostatic Features of Dengue Virus Capsid Assembly

چکیده

Dengue virus causes serious diseases and deaths in the world. Understanding fundamental mechanisms of dengue is highly demanded to develop treatments for caused diseases. Here, we present a computational work which focused on stability viral capsid. The interactions among E proteins capsid were studied using several approaches. It was found that electrostatic distribution single protein monomer inhomogeneous, makes an strongly binding with another protein. This reason why all form homodimers as basic units whole capsids. pKa calculations demonstrated folding energy low stable range pH 6–10, different from many other are at certain pH. dependence homodimer shows independent when also 6–10. finding implies can survive wide pH, explain so widely distributed world spreads fast.

The thermodynamics of virus capsid assembly.

Virus capsid assembly is a critical step in the viral life cycle. The underlying basis of capsid stability is key to understanding this process. Capsid subunits interact with weak individual contact energies to form a globally stable icosahedral lattice; this structure is ideal for encapsidating the viral genome and host partners and protecting its contents upon secretion, yet the unique proper...

A theory for viral capsid assembly around electrostatic cores.

We develop equilibrium and kinetic theories that describe the assembly of viral capsid proteins on a charged central core, as seen in recent experiments in which brome mosaic virus capsids assemble around nanoparticles functionalized with polyelectrolyte. We model interactions between capsid proteins and nanoparticle surfaces as the interaction of polyelectrolyte brushes with opposite charge us...

Irreversible growth model for virus capsid assembly.

We model the spontaneous assembly of a capsid (a virus' closed outer shell) from many copies of identical units, using entirely irreversible steps and only information local to the growing edge. Our model is formulated in terms of (i) an elastic Hamiltonian with stretching and bending stiffness and a spontaneous curvature, and (ii) a set of rate constants for the addition of new units or bonds....

Bluetongue Virus Capsid Assembly and Maturation

Maturation is an intrinsic phase of the viral life cycle and is often intertwined with egress. In this review we focus on orbivirus maturation by using Bluetongue virus (BTV) as a representative. BTV, a member of the genus Orbivirus within the family Reoviridae, has over the last three decades been subjected to intense molecular study and is thus one of the best understood viruses. BTV is a non...

Unique features of hepatitis C virus capsid formation revealed by de novo cell-free assembly.

The assembly of hepatitis C virus (HCV) is poorly understood, largely due to the lack of mammalian cell culture systems that are easily manipulated and produce high titers of virus. This problem is highlighted by the inability of the recently established HCV replicon systems to support HCV capsid assembly despite high levels of structural protein synthesis. Here we demonstrate that up to 80% of...
