چکیده

Abstract Tempol (4-hydroxy-2,2,6,6-tetramethylpiperidine-1-oxyl) is a potential redox agent in cells. The present study investigated changes cellular reactive oxygen species (ROS) and glutathione (GSH) levels antioxidant enzymes, Tempol-treated Calu-6 A549 lung cancer cells, normal WI-38 VA-13 primary pulmonary fibroblasts. Results demonstrated that (0.5–4 mM) either increased or decreased general ROS cells at 48 h specifically O 2 •− these In addition, differentially altered the expression activity of enzymes such as superoxide dismutase, catalase, thioredoxin reductase1 (TrxR1) A549, Calu-6, particular, treatment TrxR1 protein 1 mM inhibited growth by about 50% but also significantly induced cell death, evidenced annexin V-positive Furthermore, down-regulation siRNA had some effect on well inhibition death -untreated doses numbers GSH-depleted both h. conclusion, various all along with an increase GSH depletion.

Cardiac and renal antioxidant enzymes and effects of tempol in hyperthyroid rats.

This study evaluated the activity of cardiac and renal antioxidant enzymes [superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GPX), and glutathione reductase (GR)] and whether chronic treatment with tempol, a cell membrane-permeable SOD mimetic, ameliorates the hypertension of hyperthyroidism. Two experiments were performed. In experiment I, the following four groups of male W...

Therapeutic Approach to Neurodegenerative Diseases by Medical Gases: Focusing on Redox Signaling and Related Antioxidant Enzymes

Oxidative stress in the central nervous system is strongly associated with neuronal cell death in the pathogenesis of several neurodegenerative diseases such as Alzheimer's disease, Parkinson's disease, Huntington's disease, and amyotrophic lateral sclerosis. In order to overcome the oxidative damage, there are some protective signaling pathways related to transcriptional upregulation of antiox...

Antioxidant enzymes and effects of tempol on the development of hypertension induced by nitric oxide inhibition.

BACKGROUND This study analyzed whether hypertension induced by N(omega)-nitro-l-arginine methyl ester (L-NAME) is associated with dysregulation of the main antioxidant enzymes (superoxide dismutase [SOD], catalase, glutathione peroxidase [GPX], and glutathione reductase [GR]) and whether chronic administration of tempol ameliorates this hypertension. METHODS Four groups of male Wistar rats we...

Antioxidant enzymes as redox-based biomarkers: a brief review

The field of redox proteomics focuses to a large extent on analyzing cysteine oxidation in proteins under different experimental conditions and states of diseases. The identification and localization of oxidized cysteines within the cellular milieu is critical for understanding the redox regulation of proteins under physiological and pathophysiological conditions, and it will in turn provide im...

Redox mechanisms in age-related lung fibrosis

Redox signaling and oxidative stress are associated with tissue fibrosis and aging. Aging is recognized as a major risk factor for fibrotic diseases involving multiple organ systems, including that of the lung. A number of oxidant generating enzymes are upregulated while antioxidant defenses are deficient with aging and cellular senescence, leading to redox imbalance and oxidative stress. Howev...
