TCF-1 dictates lung tissue resident helper T cell and memory B cell development during flu

چکیده

Abstract Influenza (flu) infections are a significant cause of annual human morbidity and mortality. The efficacy seasonal flu vaccines depends on the accuracy prediction circulating strains for upcoming season. To develop that broadly effective more sustainable, induction cross-reactive memory immunity is desirable. Flu infection induces tissue resident T (Trm) B (Brm) cells which rapidly respond to reinfection. Tissue helper (Trh) population follicular (Tfh)-like CD4 +Trm promote development Brm cell responses following infection. transcription factor TCF-1, encoded by Tcf7, critical in both Tfh viral infections. However, role TCF-1 Trh has not been investigated. Utilizing Tcf7 fl/flCD4 Creconditional knockout mouse model conjunction with intravascular labeling influenza A peptide MHC Class II tetramers, we found required differentiation flu-specific cells, as well optimal Most significantly, upon heterosubtypic reinfection, fl/flcontrol mice expanded while it became undetectable Cremice, coinciding severely impaired lung germinal center CD138 +plasma response. Together, our data indicates response This work implications universal vaccines.

Local antigen in nonlymphoid tissue promotes resident memory CD8+ T cell formation during viral infection

Tissue-resident memory (Trm) CD8(+) T cells are functionally distinct from their circulating counterparts and are potent mediators of host protection against reinfection. Whether local recognition of antigen in nonlymphoid tissues during infection can impact the formation of Trm populations remains unresolved. Using skin infections with vaccinia virus (VacV)-expressing model antigens, we found ...

Maturation of Lymphocyte Immunophenotypes and Memory T Helper Cell Differentiation During Development in Mice

The goal of this study was to systematically investigate the ontogeny of lymphoid populations throughout postnatal development. In CD-1 mice, peak lymphocyte numbers occurred in blood on postnatal day 10 (d10) including those for natural killers (NK1.1), B cells (CD19), T helper (CD3CD4), naïve T helper (CD4CD62LposCD44low), memory T helper (CD4CD62LnegCD44high), and T cytotoxic (CD3CD8) cells....

Differential macrophage requirements for T helper cell and T helper cell-induced B lymphocyte proliferation

Major histocompatibility complex-restricted helper T cell clones against "minor" antigens expressed on B cell and macrophage surfaces, when confronted with appropriate T cell-depleted spleen cells, are induced to proliferation and, in turn, activate "target-responder" B cells to polyclonal growth and maturation. Irradiation of helper cell populations, however, demonstrates that their effector f...

Regulation of DNA methylation dictates Cd4 gene expression during development of helper and cytotoxic T cell lineages

During development, progenitor cells with binary potential give rise to daughter cells that have distinct functions. Heritable epigenetic mechanisms then lock in gene expression programs that define lineage identity. Cd4 regulation in helper and cytotoxic T cells exemplifies this process, with enhancerand silencer-regulated establishment of epigenetic memories for stable gene expression and rep...

Fibroblast and T cells conditioned media induce maturation dendritic cell and promote T helper immune response

Dendritic cells (DCs) induce pathogen-specific T cell responses. We comprehensively studied the effects of addition of maturation stimulus, fibroblasts (fibroblast conditioned medium), PHA activated T cells (T cell conditioned medium), and mixture of fibroblast & PHA activated T cells (FCM-TCCM) conditioned media on maturation of DCs. Monocytes were cultured with GM-CSF and IL-4 for five days. ...
