Targeting Ion Channel TRPM4
نویسندگان
چکیده
The transient receptor potential melastatin 4 (TRPM4) ion channel is ubiquitously expressed. Dysregulation and/or functional mutations of TRPM4 lead to several diseases. Within our studies, we screened for inhibitors and identified small molecules that block in the low µM range. Furthermore, investigated pathophysiology cardiac conditions, immune diseases cancer using these novel inhibitors, molecular biology techniques assays.

Novel screening techniques for ion channel targeting drugs
Ion channels are integral membrane proteins that regulate the flux of ions across the cell membrane. They are involved in nearly all physiological processes, and malfunction of ion channels has been linked to many diseases. Until recently, high-throughput screening of ion channels was limited to indirect, e.g. fluorescence-based, readout technologies. In the past years, direct label-free biophy...
متن کاملThe Sur1-Trpm4 Channel in Spinal Cord Injury.
Spinal cord injury (SCI) is a major unsolved challenge in medicine. Impact trauma to the spinal cord shears blood vessels, causing an immediate 'primary hemorrhage'. During the hours following trauma, the region of hemorrhage enlarges progressively, with delayed or 'secondary hemorrhage' adding to the primary hemorrhage, and effectively doubling its volume. The process responsible for the secon...
متن کاملChanging Taste by Targeting the Ion Channel TRPM5
The taste system has evolved to make nutritious foods attractive and toxic substances aversive so as to achieve a maximal health benefit for the organism. However, evolution has not kept up with changes in human diet and lifestyle, including the development of life-saving, but bitter tasting pharmaceuticals, and the over abundance of high calorie, sweet tasting, food. The recent identification ...
متن کاملVoltage dependence of the Ca2+-activated cation channel TRPM4.
TRPM4 is a Ca2+-activated but Ca2+-impermeable cation channel. An increase of [Ca2+]i induces activation and subsequent reduction of currents through TRPM4 channels. This inactivation is strikingly decreased in cell-free patches. In whole cell and cell-free configuration, currents through TRPM4 deactivate rapidly at negative potentials. At positive potentials, currents are much larger and activ...
متن کاملPhysiological roles of the TRPM4 channel extracted from background currents.
Calcium-activated nonselective cationic currents have been known for 30 years, but their physiological implications have remained unresolved until the recent cloning of the TRPM4 ion channel. Since then, TRPM4 has been identified as a key modulator of numerous calcium-dependent mechanisms such as the immune response, insulin secretion, cerebral artery constriction, respiratory rhythm, and cardi...
