Application of polymers in nucleic acid delivery
Gene therapy and immunotherapy are powerful techniques in the treatment of many life threatening diseases. The major challenge in these therapies is to seek a safe and efficient delivery carrier for gene and antigen materials. Carriers are designed to protect these molecules from degradation, improve their stability and facilitate the delivery of them to the site of action. This research study ...
متن کاملCationic lipids and polymers for gene delivery
Gene therapy, aiming to eradicate causes rather than symptoms of diseases, is believed to be the therapy of the future. A new promising area for the application of gene therapy is the emerging field of regenerative medicine, where gene delivery can be used to enhance or modify cell functions in an organ/tissue or in engineered substitutes. The aim of gene therapy is to achieve the expression of...
متن کاملCationic liposome-nucleic acid complexes for gene delivery and gene silencing.
Cationic liposomes (CLs) are studied worldwide as carriers of DNA and short interfering RNA (siRNA) for gene delivery and gene silencing, and related clinical trials are ongoing. Optimization of transfection efficiency and silencing efficiency by cationic liposome carriers requires a comprehensive understanding of the structures of CL-nucleic acid complexes and the nature of their interactions ...
متن کاملCatch and Release: Photocleavable Cationic Diblock Copolymers as a Potential Platform for Nucleic Acid Delivery.
Binding interactions between DNA and cationic carriers must be sufficiently strong to prevent nuclease-mediated degradation, yet weak enough to permit transcription. We demonstrate cationic diblock copolymers containing PEG and o-nitrobenzyl moieties that facilitated tailorable DNA complexation and light-activated release. This design unlocks a new approach to advance non-viral gene packaging.
متن کاملCationic gold microparticles for biolistic delivery of nucleic acids.
Here we report preparation and properties of positively charged gold microparticles, and their use for biolistic DNA delivery. Micron-sized gold microparticles were modified by building self-assembling polyethylenimine monolayers on their surfaces, which enabled their electrostatic interaction with negatively charged molecules such as DNA. One milligram of the surface-modified microparticles wa...
