چکیده
Abstract Tumor-infiltrating cytotoxic CD8 +T cells frequently acquire an altered state of differentiation referred to as “exhaustion”, which prevents control tumor growth. We study hetIL-15, a cytokine that stimulates the generation, proliferation and function NK cells. The objectives this was assess effects hetIL-15 immunotherapy in TNBC tumors, evaluate metabolic profile tumor-infiltrating T any potential synergy with Fatty Acid Metabolism Modulator (FAMM). Using EO771 orthotopic breast cancer model we monitored efficacy locoregional administration combination FAMM. effect treatment on growth, survival mice status immune using Seahorse flow cytometry. resulted complete regression 40% treated animals increased survival. infiltrating were tumors showed enhanced activation proliferation. Their flux analysis confirmed robust rise Oxygen Consumption Rate (OCR) substantial increase spare respiratory capacity. Combined FAMM mitochondrial function, FA uptake OCR. In addition, therapy statistically significant growth delay eradication 85% mice. Our results indicate reprogramming tumor-specific might represent strategy promote metabolically hostile Tumor Microenvironment (TME), enhancing clinical immunotherapy.

Fatty acid metabolism in breast cancer subtypes
Dysregulation of fatty acid metabolism is recognized as a component of malignant transformation in many different cancers, including breast; yet the potential for targeting this pathway for prevention and/or treatment of cancer remains unrealized. Evidence indicates that proteins involved in both synthesis and oxidation of fatty acids play a pivotal role in the proliferation, migration and inva...

IL-12 is a potent antitumor cytokine that exhibits significant clinical toxicities after systemic administration. We hypothesized that intratumoral (i.t.) administration of IL-12 coformulated with the biodegradable polysaccharide chitosan could enhance the antitumor activity of IL-12 while limiting its systemic toxicity. Noninvasive imaging studies monitored local retention of IL-12, with and w...

To establish whether insulin resistance and/or postprandial fatty acid metabolism might contribute to familial combined hyperlipidemia (FCH) we have examined parameters of insulin resistance and lipid metabolism in six FCH kindreds. Probands and relatives (n = 56) were divided into three tertiles on the basis of fasting plasma triglycerides (TG). Individuals in the highest tertile (TG > 2.5 mM;...
متن کاملRegulation of Elovl and fatty acid metabolism
Fatty acids are important regulators in the control of mammalian energy homeostasis. They are ingested in the diet but a significant amount are also endogenously produced by de novo lipogenesis. Fatty acid elongation beyond 16 carbons (palmitic acid) can occur to generate very long chain fatty acids (VLCFA), a process that is initiated by the rate-limiting condensation reaction. To date, six ma...

The mechanism of the biological oxidation and synthesis of fatty acids has largely been elucidated during the past few years through work in several laboratories (l-7). This work resulted in the complete confirmation of Knoop’s B oxidation theory and of the main conclusions derived from the early work of Dakin and Embden. Perhaps one of the most important aspects of the recent work is the demon...
