چکیده
Peptoids are a diverse family of sequence-defined oligomers N-substituted glycine monomers, that can be readily accessed by the solid-phase submonomer synthesis method. Due to versatility and efficiency this chemistry, easy access hundreds potential there is an enormous sequence space explored. This has enabled researchers from many different fields custom-design peptoid sequences tailored wide variety problems in biomedicine, nanoscience polymer science. Here we provide detailed protocols for peptoids, using optimized performed non-chemists. The method fully compatible with Fmoc-peptide conditions, so automated on existing peptide synthesizers provided here. Although peptoids well established, special considerations required order most useful desirable sidechains. methods include amino-acid-like side chains, some important non-natural monomer classes, as creation conjugates peptide-peptoid hybrids.

The enantiorecognition ability of oligomeric N-substituted glycines or "peptoids" with α-chiral, aromatic side chains was investigated by HPLC and (1)H NMR studies.
متن کاملIncorporation of unprotected heterocyclic side chains into peptoid oligomers via solid-phase submonomer synthesis.
Peptoids (N-substituted glycines) are an important class of biomimetic oligomers that have made a significant impact in the areas of combinatorial drug discovery, gene therapy, drug delivery, and biopolymer folding in recent years. Sequence-specific peptoid oligomers are easily assembled from primary amines by the solid-phase submonomer method. However, most amines that contain heterocyclic nit...
متن کاملIncorporation of chemoselective functionalities into peptoids via solid-phase submonomer synthesis.
A simple route to the introduction of a number of chemoselective functional groups into peptoids (oligo(N-substituted glycines)) by an extension of the standard solid-phase submonomer method is reported. The following groups were introduced: aminooxyacetamide, N-(carbamoylmethyl)acetohydrazide, mercaptoacetamide, 2-pyridinesulfenylmercaptoacetamide, and aldehyde-terminated peptoids. The method ...
متن کاملCis-trans amide bond rotamers in β-peptoids and peptoids: evaluation of stereoelectronic effects in backbone and side chains.
Non-natural peptide analogs have significant potential for the development of new materials and pharmacologically active ligands. One such architecture, the β-peptoids (N-alkyl-β-alanines), has found use in a variety of biologically active compounds but has been sparsely studied with respect to folding propensity. Thus, we here report an investigation of the effect of structural variations on t...
متن کاملSolid-phase Submonomer Synthesis of Peptoid Polymers and their Self-Assembly into Highly-Ordered Nanosheets
Peptoids are a novel class of biomimetic, non-natural, sequence-specific heteropolymers that resist proteolysis, exhibit potent biological activity, and fold into higher order nanostructures. Structurally similar to peptides, peptoids are poly N-substituted glycines, where the side chains are attached to the nitrogen rather than the alpha-carbon. Their ease of synthesis and structural diversity...
