Strategies for Recovery, Purification and Quantification of Torularhodin Produced by Rhodotorula mucilaginosa Using Different Carbon Sources

Torularhodin is a fungus-derived carotenoid, and the lack of downstream processing torularhodin still challenge for its large-scale production. To support industrial production torularhodin, this work initially evaluated efficiency carotenoid release from Rhodotorula mucilaginosa using thermal acid treatment, saponification ultrasound-assisted enzymatic lysis. Based on polarity, was then purified methanol/acetone/hexane (2/2/1, v/v/v) solution eluting silica cartridge. Thermal treatment considered most appropriate method total recovery. The highest content 121.3 ± 7.0 μg/g dry cell weight 63.0 6.1% (50.5 3.0 in total) recovered after purification. fast quantify extracted yeast, absorption coefficient (E1cm1% = 3342) dissolved chloroform assayed. With developed strategy recovery, purification quantification, potential yeast to produce xylose glycerol further evaluated. It found that carbon sources may influence proportion carotenoids but dominant pigment. results obtained study identified feasibility sustainable with high purity.
