Stoichiometry and role of GTP hydrolysis in bovine neurotubule assembly.
A method is given for preparing tubulin with 1 mol of exchangeably bound [gamma-32P]GTP/mol of 6 S dimer. Bovine tubulin is shown to hydrolyze 1 mol of GTP/mol of 6 S dimer added to assembling microtubules at 37 degrees. Hydrolysis and assembly occur at the same rate and to the same extent. When microtubule-associated proteins (MAPs) are removed, both hydrolysis and assembly fail to occur. Read...
متن کاملIncorporation of tubulin subunits into dimers requires GTP hydrolysis.
A toroid multisubunit complex of 800-900 kDa has been implicated in assisting protein folding of at least two cytoplasmic proteins, actin and tubulin. This process is dependent on the presence of magnesium ions and ATP hydrolysis. In vitro translation of cDNAs encoding different alpha- and beta-tubulin isotypes also gives rise to the formation of complexes of about 300 kDa. These complexes have...
متن کاملTubulin GTP hydrolysis influences the structure, mechanical properties, and kinesin-driven transport of microtubules.
Tubulin is a GTPase that hydrolyzes its bound nucleotide triphosphate after it becomes incorporated into a microtubule. The only known consequence of nucleotide hydrolysis is that it increases the dissociation rate of tubulin from the end of the microtubule by 2 orders of magnitude. In this study, we investigated whether microtubules composed of tubulin-GMPCPP (guanylyl alpha,beta-methylenediph...
متن کاملHigh-Resolution Microtubule Structures Reveal the Structural Transitions in αβ-Tubulin upon GTP Hydrolysis
Dynamic instability, the stochastic switching between growth and shrinkage, is essential for microtubule function. This behavior is driven by GTP hydrolysis in the microtubule lattice and is inhibited by anticancer agents like Taxol. We provide insight into the mechanism of dynamic instability, based on high-resolution cryo-EM structures (4.7-5.6 Å) of dynamic microtubules and microtubules stab...

Institute of Molecular Biology proteins dimerize through their N-terminal domains, proCopenhagen University ducing a “stalk” structure on the 50S subunit with a Sølvgade 83 flexible hinge region that probably allows the C-terminal DK1307-Copenhagen K domains of the dimers to move relative to one another Denmark and to the ribosomal surface (Traut et al., 1995). The rRNA region is also involved,...
