چکیده
Abstract Glioblastoma is a devastating disease with an overall median survival of 8 months from diagnosis. The majority patients die tumor relapse in close proximity the resected primary tumor. intensely researched but most studies have been performed on tissues and cultures derived bulk while it remaining edge cells that cause lethality. Investigations are rare few experimental models exist. Here we established analyzed series matched cell outer six IDH wildtype glioblastoma purpose to understand biology. Tumor samples were guided by 5-ALA fluorescence using neuro-navigation stringent procedures not contaminate cells. First fluorescent tissue. After removal all areas careful irrigation cavity sample was 1-2 cm outside border non-eloquent area. Following dissociation used sphere assays for explantation. There significant difference self-renewal across between cultures, line results acute cells, suggesting maintenance properties cultures. Invasion analysis showed reverse strengthening general functional distinction patients. To investigate molecular basis our findings whole exome sequencing (WES) combined single RNA- ATAC-sequencing (10X Multiome). Analyses ongoing WES data does support genetic causes their differences 10X Multiome indicate epigenetic regulation may underlie different

Glioblastoma and stem cells Minireview
This review presents compelling evidence that human glioblastoma is a heterogenous tumor composed from tumor cells and small portion of cancer stem cells – tumor-initiating cells, which have a high tumorigenic potential and a low proliferation rate. Glioma cancer stem cells are phenotypically similar to the normal stem cells, they express CD133 gene and other genes characteristic of neural stem...
متن کاملModeling Microenvironmental Regulation of Glioblastoma Stem Cells: A Biomaterials Perspective
Following diagnosis of a glioblastoma (GBM) brain tumor, surgical resection, chemotherapy, and radiation together yield a median patient survival of only 15 months. Importantly, standard treatments fail to address the dynamic regulation of the brain tumor microenvironment that actively supports tumor progression and treatment resistance. It is becoming increasingly recognized that specialized n...
متن کاملGlioblastoma Multiforme Stem Cells
Glioblastoma multiforme (GBM) is an aggressive, malignant, and lethal brain tumor, resistant to all current forms of treatment. The rapidly emerging focus on cancer stem cells embodies a paradigm shift in our understanding of tumor pathogenesis, while the development of powerful genome-wide screening techniques has provided cause for optimism related to the development of more reliable therapie...
متن کاملO27: Interaction of Cancer Stem Cells and Microglia in Glioblastoma Multiforme
Malignant gliomas are highly invasive brain tumors with the occurrence of multiple microglia/macrophages in the tumor microenvironment. Macrophages/microglia that found in glioma microenvironment, as tumor-infiltrating immune cells, can play a harmful role in tumor progression. In addition, glioblastoma multiforme (GBM) contains multiple aberrant differentiation and tumorigenic cancer stem cell...
متن کاملCancer stem cells in glioblastoma.
Tissues with defined cellular hierarchies in development and homeostasis give rise to tumors with cellular hierarchies, suggesting that tumors recapitulate specific tissues and mimic their origins. Glioblastoma (GBM) is the most prevalent and malignant primary brain tumor and contains self-renewing, tumorigenic cancer stem cells (CSCs) that contribute to tumor initiation and therapeutic resista...
