The Vero cell is the continuous line used as a substrate for many viral vaccines manufacturing including Rabies vaccine. Pitman and PV-11 strains of rabies virus are commonly production vaccines. In our study PV - 11 strain seed derived inactivated Anti Vaccines (Lyophilized). working bank lot were prepared tested its sterility mycoplasma contamination. this roller bottle method micro carrier (cytodex-1) based bioreactor culturing standardized with MEM Eagles RPMI 1640 medium. outcome two methods compared ,to find out medium which can give higher concentration cells to get titre in mass production. quantities obtained from passage 144 148 assessed using haemocytometer count procedure. quantity per ranged 222.82x106 236.67x106 227.80x106 230.66x106 average bottles 1.12 x 106 ml culture. 1.69 1.75 million / culture by 1.67 1.70 yield vero x106 It estimated that about 35% increase system than method. 0.3 MOI (Multiplicity Infection) was kept constant infection confluent monolayered both Four harvests collected at three day intervals replenished fresh each time. titres harvest quantified in-house RT-PCR method; yielded log 6.044 7.106 6.559 7.216 particles. observed more
