چکیده
ObjectiveTo explore the link between sphingosine 1-phosphate (S1P) signaling and leiomyoma possible S1P cross-talk with fibrotic effect of activin A.DesignCase-control laboratory study.SettingUniversity institute university hospital.Patient(s)Patients uterine fibroids (n = 26).Interventions(s)Tissue specimens normal myometrium were obtained from patients undergoing myomectomy or total hysterectomy.Main Outcome Measure(s)Expression mRNA levels enzyme involved in metabolism, receptors, transporter Spns2 was evaluated matched leiomyoma/myometrium cell populations. The effects inhibition metabolism on A–induced action lines.Result(s)The expression enzymes responsible for formation, kinase (SK) 1 2, S1P2, S1P3, S1P5 receptors significantly augmented leiomyomas compared adjacent myometrium. In cells, but not myometrial A increased SK1, SK2, S1P2. profibrotic abolished when SK1/2 inhibited S1P2/3 blocked. Finally, by itself markers (fibronectin, collagen 1A1) cells. Conclusion(s)This study shows that is dysregulated fibrosis, opening new perspectives fibroid treatment. To A. Case-control study. Sphingosine-1-phosphate signaling in endothelial activation.
The signaling and functions of the Endothelial Differentiation Gene (EDG) family of G protein-coupled receptors have been extensively elucidated. All the members of EDG family were shown to be receptors for lysosphingolipids or lysophospholipids. EDG-1, the prototype of EDG family receptors, is a high affinity receptor for serum-borne bioactive lipid, Sphingosine-1-phosphate (S1P). S1P, secrete...

BACKGROUND The accumulation of beta amyloid (Aβ) peptides, a hallmark of Alzheimer's disease (AD) is related to mechanisms leading to neurodegeneration. Among its pleiotropic cellular effects, Aβ accumulation has been associated with a deregulation of sphingolipid metabolism. Sphingosine 1-phosphate (S1P) derived from sphingosine is emerging as a critical lipid mediator regulating various biolo...
متن کاملImplication of Ceramide, Ceramide 1-Phosphate and Sphingosine 1-Phosphate in Tumorigenesis
In the last two decades there has been considerable progress in our understanding of the role of sphingolipids in controlling signal transduction processes, particularly in the mechanisms leading to regulation of cell growth and death. Ceramide is a well-characterized sphingolipid metabolite and second messenger that can be produced by cancer cells in response to a variety of stimuli, including...

Besides their structural role, sphingolipids including Sphingosine-1phosphate (S1P) have drawn attention as bioactive sphingolipid signaling molecules involved in the regulation of several processes, including cell growth, differentiation, survival, migration or immune response. It is now well established that S1P is involved in the onset or the progression of pathological conditions such as ca...
متن کاملSphingosine 1-Phosphate Receptor 1 Signaling in Mammalian Cells.
The bioactive lipid, sphingosine 1-phosphate (S1P) binds to a family of G protein-coupled receptors, termed S1P₁-S1P₅. These receptors function in, for example, the cardiovascular system to regulate vascular barrier integrity and tone, the nervous system to regulate neuronal differentiation, myelination and oligodendrocyte/glial cell survival and the immune system to regulate T- and B-cell subs...
