Spectral studies of lactose repressor protein modified with nitrophenol reporter groups.
The spectral characteristics of lactose repressor protein modified with two nitrophenol reporter groups have been determined. Reaction of Z-chloromercuri-4nitrophenol with repressor increases the apparent pK of the nitrophenol from 6.75 to 8.3 and alters the wavelength of maximum absorption. Subsequent binding of inducer shifts the apparent pK toward slightly more acid pH. The pK values of the ...
متن کاملChemical modification of lactose repressor protein using N-substituted maleimides.
Lactose repressor protein has been modified with N-ethylmaleimide, two N-maleimide spin labels, and an N-maleimide fluorophore. The reaction with repressor cysteine residues has been characterized. Approximately 2 of the 3 eq of cysteine/repressor monomer are reactive toward these reagents. Repressor cysteines are reactive toward these reagents in the order cysteine 140 greater than or equal to...
متن کاملSpectral triples of weighted groups
We study spectral triples on (weighted) groups and consider functors between the categories of weighted groups and spectral triples. We study the properties of weights and the corresponding functor for spectral triples coming from discrete weighted groups.
متن کاملProtein and Lactose Separation by Modified Ultrafiltration Membrane using Layer by Layer Technique
Layer-by-Layer (LbL) is a method which can be used for nanoscale coating and surface functionalization of a material. LbL technique mainly uses the electrostatic attracting between charged materials (polyelectrolytes, nanoparticles, etc.) and an oppositely charged surface. In this study, protein separation (BSA) from lactose solution was carried out using the LbL self-assembly method, which was...
متن کاملIn vitro interaction of the Escherichia coli cyclic AMP receptor protein with the lactose repressor.
Sedimentation equilibrium studies show that the Escherichia coli cyclic AMP receptor protein (CAP) and lactose repressor associate to form a 2:1 complex in vitro. This is, to our knowledge, the first demonstration of a direct interaction of these proteins in the absence of DNA. No 1:1 complex was detected over a wide range of CAP concentrations, suggesting that binding is highly cooperative. Co...
