Specific and sensitive plate assay for bacterial lipases.
A plate assay to detect bacterial lipase (EC 3.1.1.3) in a medium containing trioleoylglycerol and the fluorescent dye rhodamine B is presented. Substrate hydrolysis causes the formation of orange fluorescent halos around bacterial colonies visible upon UV irradiation. The logarithm of lipase activity from cell-free culture supernatants is linearly correlated with the diameter of halos, thereby...
متن کاملPreparation of prostate specific antigen standards for immunoradiometric assay
Background: Immunoradiometric assay is one of the most common and precise methods for determination of prostate specific antigen (PSA) in clinical laboratories. Usual use of human serum in routine assays has many disadvantages such as easy contamination and precipitation, instability and unavailability. Thus in order to avoid these problems the artificial matrix was used which acts sim...
متن کامل[Spore rec-assay by dry sheet medium culture plate for bacterial counts].
A simple and rapid method for spore rec-assay by utilizing dry sheet medium culture (Compactdry TC, CTC) for determining numbers of bacteria, instead of the spore agar plate, was developed. One mL of spore suspension (2 x 10(6)/mL) of Bacillus subtilis strain M45 Rec- or H17 Rec+ was inoculated in the center of the CTC plate. In the case of metabolic activation, 1 mL of mixed solution (spore su...
متن کاملA simple microtiter plate screening assay for bacterial invasion or adherence
Many bacteriologic studies, including cellular invasion and adherence assays, require enumeration of viable organisms or colony forming units. When attempting to screen large numbers of clinical or environmental isolates or laboratoryderived mutants for differences in invasion or adherence phenotype, standard plating methods can be
متن کاملDual-substrate plate diffusion assay for proteases.
A plate diffusion assay for endopeptidases was developed. Proteases applied to plates containing 1% casein, 1% gelatin, and 1.5% agar caused distinct zones reminiscent of immunoprecipitation bands. The diameter of the zones was linearly proportional to the log of the enzyme activity applied over a range from 0.01 to greater than 100 IU/ml.
