SP5.1 Paracrine Interferon signalling between cancer associated Fibroblasts and Cancer Cells correlates with poor survival after chemotherapy in Triple Negative Breast Cancer patients

Abstract Aim To determine if expression levels of Interferon-β1 (IFNβ1) in cancer associated fibroblasts, and Myxovirus Resistance 1 (MX1) – a downstream marker interferon activity, cells would be predictive markers chemotherapy response subsequent overall survival. Methods Breast tissue was obtained from microarrays triple negative cohort. Expression IFNβ1 MX1 determined by immunohistochemistry 109 patients treated with adjuvant chemotherapy. The cohort further subdivided into claudin low high subgroups, based on immunohistochemical assessment relative claudin-3. Results fibroblasts significantly, positively tumour (Spearman’s correlation r=0.210; p=0.028). High were each significantly poorer disease-free survival (p=0.01). Dividing the claudin-low (claudin-3 negative; n=49) claudin-high positive; n=60), resulted stronger between fibroblast cell group (r=0.375; p=0.008), whereas it lost (r=0.113; p=0.389). Likewise, correlations maintained cases (p<0.05) but claudin-highs. Conclusion may value as biomarkers chemo-response breast patients. These imitate associations chemoresistance identified vitro using lines. Therefore, inhibiting paracrine cross talk-between types might viable strategy for chemo-sensitisation cancers.
