چکیده

Akebia trifoliata is a perennial woody plant with considerable potential in nutrition, food, and health, the production of seedlings high quality critical for its economic utilization. Plant regeneration through somatic embryogenesis powerful alternative propagating many species. In this study, simple practicable protocol was developed from immature seeds A. via embryogenesis, genetic stability regenerated plants also assessed. embryo (SE) induction stage, highest frequency (35.2%) observed on WPM medium containing 1.0 mg L?1 thidiazuron (TDZ) 6-benzyladenine (6-BA). The concentration 6-BA optimized at proliferation maturation induced SEs, combination 2.0 indole-3-butyric acid (IBA) 0.5 TDZ most responsive root development growth. leaf morphological characteristics greatly varied among established plants, they could be grouped into three types, namely normal type, Type ?, ??. Remarkable differences number, size, shape, color leaflets were while their ploidy level same flow cytometry analysis. ?? ? had lowest net photosynthesis rate, transpiration stomatal conductance groups, respectively, both smaller size guard cells than type. Simple sequence repeat (SSR) analysis detected that 41 bands (43.62%) different those wild, indicating degree polymorphism between regenerants donor parent. obtained might hold future improvement breeding trifoliata, serve as foundation vitro propagation germplasm preservation crop.

Akebia trifoliate (Thunb.) Koidz Seed Extract Inhibits the Proliferation of Human Hepatocellular Carcinoma Cell Lines via Inducing Endoplasmic Reticulum Stress

Akebia Fructus has long been used for hepatocellular carcinoma (HCC) in China, while the molecular mechanism remains obscure. Our recent work found that Akebia trifoliate (Thunb.) Koidz seed extract (ATSE) suppressed proliferation and induced endoplasmic reticulum (ER) stress in SMMC-7721. The present study aimed to throw more light on the mechanism. ER stress occurred after ATSE treatment in H...

Induction of Callus and Somatic Embryogenesis from Cotyledon Explants of Fagonia indica Burm.

To explore the potential for in vitro rapid regeneration of Fagonia indica Burm., cotyledon explants were obtained from 10-day old aseptically germinated seedlings that cultured on MS medium which supplemented with 1.0-5.0 µM 2,4-D. Callus was initiated from cotyledon explant on MS medium supplemented in all concentrations of 2,4-D investigated. 2,4-D was more effective than IAA and NAA for cal...

Somatic Versus Zygotic Embryogenesis: Learning from Seeds.

Plant embryogenesis is a fascinating developmental program that is very successfully established in nature in seeds. In case of in vitro somatic embryogenesis this process is subjected to several limitations such as asynchronous differentiation and further development of somatic embryos, malformations and disturbed polarity, precocious germination, lack of maturity, early loss of embryogenic po...

Callus Induction and Somatic Embryogenesis in Muscadine and Seedless Bunch Grapes (vitis) from Immature Ovule Culture

Pre-embryogenic calli were obtained from the culture of fertilized immature ovules of muscadine ( Vitis rotundifolia Michx.) and seedless bunch grapes ( Vitis vinifera L.) on: 1) Murashige & Turcker (MT, 1969) basal medium supplemented with 4.4 μM 2, 4-D (2, 4-dichlorophenoxyacetic acid) and 1 μM BA (6-benzyladenine) with 6.0% (w/v) sucrose; 2) Chee and Pool (CP, 1978) basal medium supplemented...

Loss of CMD2‐mediated resistance to cassava mosaic disease in plants regenerated through somatic embryogenesis

Cassava mosaic disease (CMD) and cassava brown streak disease (CBSD) are the two most important viral diseases affecting cassava production in Africa. Three sources of resistance are employed to combat CMD: polygenic recessive resistance, termed CMD1, the dominant monogenic type, named CMD2, and the recently characterized CMD3. The farmer-preferred cultivar TME 204 carries inherent resistance t...
