Sleep and Metabolism: Implication of Lateral Hypothalamic Neurons

چکیده

During the last decade, optogenetic-based circuit mapping has become one of most common approaches to systems neuroscience, and amassing studies have expanded our understanding brain structures causally involved in regulation sleep-wake cycles. Recent imaging technologies enable functional cellular activity, from population down single-cell resolution, across a broad repertoire behaviors physiological processes, including states. This chapter summarizes experimental evidence implicating hypocretins/orexins, melanin-concentrating hormone, inhibitory neurons lateral hypothalamus (LH) forming an intricate network regulating sleep metabolism, feeding behaviors. It further confirms dual sleep-metabolic functions LH cells, sheds light on possible mechanism underlying plasticity during metabolic disorders.

Sleep-waking discharge patterns of neurons recorded in the rat perifornical lateral hypothalamic area.

The perifornical lateral hypothalamic area (PF-LHA) has been implicated in the control of several waking behaviours, including feeding, motor activity and arousal. Several cell types are located in the PF-LHA, including projection neurons that contain the hypocretin peptides (also known as orexins). Recent findings suggest that hypocretin neurons are involved in sleep-wake regulation. Loss of h...

Sleep and metabolism: role of hypothalamic neuronal circuitry.

Sleep and metabolism are intertwined physiologically and behaviorally, but the neural systems underlying their coordination are still poorly understood. The hypothalamus is likely to play a major role in the regulation sleep, metabolism, and their interaction. And increasing evidence suggests that hypocretin cells in the lateral hypothalamus may provide particularly important contributions. Her...

Direct innervation and modulation of orexin neurons by lateral hypothalamic LepRb neurons.

Leptin, the adipose-derived hormonal signal of body energy stores, acts via the leptin receptor (LepRb) on neurons in multiple brain regions. We previously identified LepRb neurons in the lateral hypothalamic area (LHA), which are distinct from neighboring leptin-regulated melanin-concentrating hormone (MCH)- or orexin (OX)-expressing cells. Neither the direct synaptic targets of LHA LepRb neur...

Vasopressin and oxytocin excite MCH neurons, but not other lateral hypothalamic GABA neurons.

Neurons that synthesize melanin-concentrating hormone (MCH) colocalize GABA, regulate energy homeostasis, modulate water intake, and influence anxiety, stress, and social interaction. Similarly, vasopressin and oxytocin can influence the same behaviors and states, suggesting that these neuropeptides may exert part of their effect by modulating MCH neurons. Using whole cell recording in MCH-gree...

The lateral hypothalamic area controls paradoxical (REM) sleep by means of descending projections to brainstem GABAergic neurons.

It has recently been shown that the ventrolateral part of the periaqueductal gray (VLPAG) and the adjacent dorsal deep mesencephalic nucleus (dDpMe) contain GABAergic neurons gating paradoxical sleep (PS) onset by means of their projection to the glutamatergic PS-on neurons of the sublaterodorsal tegmental nucleus (SLD). To determine the mechanisms responsible for the cessation of activity of t...
