Site specific functionalization of oligonucleotides for attaching two different reporter groups.
The synthesis of an oligonucleotide functionalized to attach two different reporter groups at specific internucleotide linkages is described. To incorporate the amine specific reporter group the internucleotide linkage is modified to phosphoramidate (N-1-aminoalkyl) and for a thiol specific reporter group the internucleotide linkage is modified to a phosphorothioate diester. The synthetic cycle...
متن کاملStrategies for Attaching Oligonucleotides to Solid Supports
While most molecular reactions involving synthetic oligonucleotides, notably PCR and DNA sequencing, are carried out in liquid phase, a rapidly growing number of important applications utilize synthetic oligonucleotides attached to solid supports. In general, the purpose for using a solid support-attached oligonucleotide is to capture something, hold on to it, enrich it, and identify and/or pur...
متن کاملStrategies for Attaching Oligonucleotides to Solid Support
Many important molecular applications, such as DNA oligonucleotide arrays, utilize synthetic oligonucleotides attached to solid supports. The most accessible approach for producing an oligonucleotide microarray is to synthesize individual oligonucleotides and subsequently immobilize them to a solid surface. For this immobilization to take place, the oligonucleotides must be modified with a func...
متن کاملAsymmetric functionalization of gold nanoparticles with oligonucleotides.
Gold nanoparticles (AuNPs) were anisotropically functionalized with two different oligonucleotide sequences using magnetic microparticles as geometric restriction templates for site-selective enzymatic extension of particle-bound oligonucleotides. The divalent linking capability of the resulting AuNPs allowed for the design and programmable assembly of discrete nanoparticle heterostructures.
