Single or Divided Administration of Cisplatin Can Induce Inflammation and Oxidative Stress in Male Sprague-Dawley Rats

BACKGROUND: Cisplatin is one of the most potent chemotherapy drugs to treat various types cancer, however use cisplatin has some adverse effect, such as increase oxidative stress and inflammation by malondialdehyde (MDA) nuclear factor kappa B (NF-kB) activation. Since dosing critical, we observed effect single multiple doses injection on rats’ level.METHODS: Total 27 male Sprague-Dawley rats were divided into 9 sub-groups, each consisted 3 rats. The baseline sub-group received no treatments; Group 1 (sub-group 1.1, 1.2, 1.3, 1.4) administered dose 5 mg/kg BW/intravenously (i.v) cisplatin; 2 2.1, 2.2, 2.3, 2.4) given 0.2 BW/i.v twice a week for two months. Rats their body weight, NF-kB, MDA level based assigned group. RESULTS: Body weight loss was in 1st after treatment 1, 7th 2. showed increasing NF-kB since observation, which 5th week, respectively. levels also significantly both groups every observation (p<0.05).CONCLUSION: either or can induce thus decrease weight. Safety of red ginseng oil for single oral administration in Sprague–Dawley rats

The single oral administration of red ginseng oil (5000 mg/kg) to Sprague-Dawley rats induced no changes in behavioral patterns, clinical signs, and body weight, and hepatotoxicity parameters such as aspartate aminotransferase and alanine aminotransferase for 14 d.
