Single-Cell RNA Sequencing of Donor-Reactive T Cells Reveals Role of Apoptosis in Donor-Specific Hyporesponsiveness of Kidney Transplant Recipients
نویسندگان
چکیده
After kidney transplantation (KT), donor-specific hyporesponsiveness (DSH) of recipient T cells develops over time. Recently, apoptosis was identified as a possible underlying mechanism. In this study, both transcriptomic profiles and complete V(D)J variable regions TR transcripts from individual alloreactive transplant recipients were determined with single-cell RNA sequencing. Alloreactive by CD137 expression after stimulation peripheral blood mononuclear obtained KT (N = 7) prior to 3–5 years their donor or third party control. The sorted, sequenced the transcriptome cell receptor analyzed using unsupervised clustering. retain highly polyclonal Cell Receptor Alpha/Beta repertoire Post transplantation, donor-reactive CD4+ had specific downregulation genes involved in cytokine-mediated pathways apoptosis. CD8+ profile did not change significantly Single-cell profiling shows that activated pro-apoptotic clones are preferentially lost stable recipients.

Effect of donor-specific antibodies and panel reactive antibodies in living donor liver transplant recipients
PURPOSE Preformed circulating donor-specific antibodies (DSAs) immunologically challenge vascular endothelium and the bile duct. However, the liver is an immune-tolerant organ and can avoid immunological challenges. This study was undertaken to analyze the effects of DSAs after adult living donor liver transplantation (LDLT). METHODS We retrospectively reviewed 219 LDLT patients' records trea...
متن کاملDonor-Derived Myeloid Sarcoma in Two Kidney Transplant Recipients from a Single Donor
We report the rare occurrence of donor-derived myeloid sarcoma in two kidney transplant patients who received organs from a single deceased donor. There was no evidence of preexisting hematologic malignancy in the donor at the time of organ recovery. Both recipients developed leukemic involvement that appeared to be limited to the transplanted organ. Fluorescence in situ hybridization (FISH) an...
متن کاملAortic stiffness of kidney transplant recipients correlates with donor age.
Increased aortic stiffness is a major factor responsible for the high cardiovascular mortality in patients with end-stage renal disease, but the impact of kidney transplantation on recipient aortic stiffness remains poorly defined. The use of expanded-criteria kidney donors is associated with decreased recipient survival compared with the use of standard-criteria donors, although the underlying...
متن کاملAnti-HLA Antibodies and Kidney Allograft Outcomes in Recipients with Donor Bone Marrow Cell Infusion
Background: Anti-HLA-antibodies are known to affect the allograft survival in transplant recipient patients. Objective: The aim of this study was to evaluate the association between anti-HLA antibodies and kidney allograft outcomes, particularly in recipients with concur-rent donor bone marrow cell infusion (DBMI). Methods: Between June 2006 and May 2007, forty living unrelated donor kidney tra...
متن کاملComment on "Tracking donor-reactive T cells: Evidence for clonal deletion in tolerant kidney transplant patients".
T cell responses to allogeneic major histocompatibility complex antigens present a formidable barrier to organ transplantation, necessitating long-term immunosuppression to minimize rejection. Chronic rejection and drug-induced morbidities are major limitations that could be overcome by allograft tolerance induction. Tolerance was first intentionally induced in humans via combined kidney and bo...
