Simvastatin-romidepsin combination kills bladder cancer cells synergistically

چکیده
The HMG-CoA reductase inhibitor simvastatin activates AMP-activated protein kinase (AMPK) and thereby induces histone acetylation. We postulated that combining with the deacetylase (HDAC) romidepsin would kill bladder cancer cells by inducing acetylation cooperatively. combination of induced robust apoptosis killed synergistically. In murine subcutaneous tumor models using MBT-2 cells, a 15-day treatment 0.5 mg/kg 15 was well tolerated inhibited growth significantly. Mechanistically, activating AMPK. also decreased expression HDACs, thus further promoting This AMPK activation essential for combination's action because compound C, an inhibitor, suppressed combination-induced ability to induce apoptosis. found increased peroxisome proliferator-activated receptor (PPAR) γ, leading reactive oxygen species production. Furthermore, endoplasmic reticulum (ER) stress this ER shown be associated acetylation, playing important role in action. Our study suggests there is positive feedback cycle between induction PPARγ expression.

Combination of metformin and phenformin synergistically inhibits proliferation and hTERT expression in human breast cancer cells
Objective(s): Breast cancer remains a global challenge, and further chemopreventive therapies are still immediately required. Emerging evidence has revealed the potent anti-cancer effects of biguanides, Metformin (MET) and phenformin (PHE). Thus, to explore an efficient chemopreventive strategy for breast cancer, the antiproliferative effects of the combination of MET and PHE against breast can...
متن کاملSimultaneous inhibition of deubiquitinating enzymes (DUBs) and autophagy synergistically kills breast cancer cells
Breast cancer is one of the leading causes of cancer death among women in the United States. Patients expressing the estrogen and progesterone receptor (ER and PR) and human epidermal growth factor 2 (HER-2) tumor markers have favorable prognosis and efficacious therapeutic options. In contrast, tumors that are negative for these markers (triple-negative) have a disproportionate share of morbid...
متن کاملMetformin kills and radiosensitizes cancer cells and preferentially kills cancer stem cells
UNLABELLED The anti-cancer effects of metformin, the most widely used drug for type 2 diabetes, alone or in combination with ionizing radiation were studied with MCF-7 human breast cancer cells and FSaII mouse fibrosarcoma cells. Clinically achievable concentrations of metformin caused significant clonogenic death in cancer cells. Importantly, metformin was preferentially cytotoxic to cancer st...

doxorubicin is a broad spectrum antibiotic used in the treatment of cancers. its dose dependent cardiotoxicity is the most serious side effect causing withdrawal of drug from hard chemotherapeutic regimen. statins are shown to be cytotoxic in concentrations higher than the effective doses for the treatment of hypercholesterolemia (40 mg/day). co-administration of statins and chemotherapeutic ag...
