Shifts in DNA yield and biological community composition in stored sediment: implications for paleogenomic studies

چکیده
Lake sediments hold a wealth of information from past environments that is highly valuable for paleolimnological reconstructions. These studies increasingly apply modern molecular tools targeting sedimentary DNA (sedDNA). However, sediment core sampling can be logistically difficult, making immediate subsampling sedDNA challenging. Sediment cores are often refrigerated (4 °C) weeks or months before subsampling. We investigated the impact storage time on changes in (purified as cell lysate) concentrations and shifts biological communities following lake surface at 4 °C up to 24 weeks. samples (~ 0.22 g, triplicate per point) were spiked with purified (100 200 ng) lysate brackish water cyanobacterium produces cyanotoxin nodularin non-spiked. Samples analysed every 1–4 over 24-week period. Droplet digital PCR showed no significant decrease target gene ( synthetase – subunit F; ndaF ) period DNA, while copy number decreased by more than half lysate-spiked samples. There was change bacteria eukaryotic community composition assessed using metabarcoding. Amongst bacteria, cyanobacterial signal became negligible after 5 Proteobacteria increased. In community, Cercozoa dominant 6 data demonstrate yields shift significantly when stored chilled This highlights need rapid appropriate paleogenomic studies.

simulation and experimental studies for prediction mineral scale formation in oil field during mixing of injection and formation water
abstract: mineral scaling in oil and gas production equipment is one of the most important problem that occurs while water injection and it has been recognized to be a major operational problem. the incompatibility between injected and formation waters may result in inorganic scale precipitation in the equipment and reservoir and then reduction of oil production rate and water injection rate. ...
Shifts in coastal sediment oxygenation cause pronounced changes in microbial community composition and associated metabolism
BACKGROUND A key characteristic of eutrophication in coastal seas is the expansion of hypoxic bottom waters, often referred to as 'dead zones'. One proposed remediation strategy for coastal dead zones in the Baltic Sea is to mix the water column using pump stations, circulating oxygenated water to the sea bottom. Although microbial metabolism in the sediment surface is recognized as key in regu...

this study intends to investigate samuel beckett’s waiting for godot and endgame under the lacanian psychoanalysis. it begins by explaining the most important concepts of lacanian psychoanalysis. the beckettian characters are studied regarding their state of unconscious, and not the state of consciousness as is common in most beckett studies. according to lacan, language plays the sole role in ...

[1] Mechanistic models for sediment transport on hillslopes are needed for applications ranging from landscape evolution to debris-flow hazards. Progress has been made for soil-mantled landscapes; however, little is known about sediment production and transport in bedrock landscapes that often maintain a patchy soil mantle, even though slopes exceed the angle of repose. Herein we investigate th...
