چکیده

Rice stripe virus (RSV) is a plant virus, and its infection leads to devastating disease that often causes serious yield losses in rice-producing regions. In this study, the capsid protein (CP) gene of RSV was cloned expressed E. coli (Rosetta), purified recombinant His-CP used immunize New Zealand white rabbits . After six weeks, polyclonal antibody against (PAb-CP ) obtained from serum immunized rabbits. Using prepared PAb-CP , three serology-based detection methods were developed, which could specifically test for RSV-infected rice plants or small brown planthoppers (SBPHs). Methods combine dot-blot with enzyme-linked immunosorbent assay high-throughput also developed detect seedlings SBPHs simultaneously wild fields. The field surveys indicated incidence suburbs Yangzhou City much lower than value ten years ago. This research presents multiple methods, using can identify conditions. Our study will benefit control planting • optimal titer coat 1 × 10 5 be by serological-based methods. both SBPs. Dot blot recommended

Transcriptome Analysis of the Small Brown Planthopper, Laodelphax striatellus Carrying Rice stripe virus

Rice stripe virus (RSV), the type member of the genus Tenuivirus, transmits by the feeding behavior of small brown planthopper (SBPH), Laodelphax striatellus. To investigate the interactions between the virus and vector insect, total RNA was extracted from RSV-viruliferous SBPH (RVLS) and non-viruliferous SBPH (NVLS) adults to construct expressed sequence tag databases for comparative transcrip...

Detection of Quantitative Trait Loci (QTLs) for Resistances to Small Brown Planthopper and Rice Stripe Virus in Rice Using Recombinant Inbred Lines

Small brown planthopper (SBPH) and rice stripe virus (RSV) disease transmitted by SBPH cause serious damage to rice (Oryza sativa L.) in China. In the present study, we screened 312 rice accessions for resistance to SBPH. The indica variety, N22, is highly resistant to SBPH. One hundred and eighty two recombinant inbred lines (RILs) derived from a cross of N22 and the highly susceptible variety...

Viruliferous rate of small brown planthopper is a good indicator of rice stripe disease epidemics

Rice stripe virus (RSV), its vector insect (small brown planthopper, SBPH) and climatic conditions in Jiangsu, China were monitored between 2002 and 2012 to determine key biotic and abiotic factors driving epidemics of the disease. Average disease severity, disease incidence and viruliferous rate of SBPH peaked in 2004 and then gradually decreased. Disease severity of RSV was positively correla...

Analysis of rice stripe virus whole-gene expression in rice and in the small brown planthopper by real-time quantitative PCR.

Rice stripe virus (RSV) is a typical member of the Tenuivirus genus, which is transmitted mainly by the small brown planthopper (SBPH) in a persistent, circulative-propagative manner. At present, expression levels of viral genes in rice and in SBPH remain unclear. In this study, we investigated RSV gene expression in RSV-infected rice tissues and in viruliferous SBPH using a sensitive and relia...

Transmission of Rice stripe virus acquired from frozen infected leaves by the small brown planthopper (Laodelphax striatellus Fallen).

Rice stripe disease, caused by Rice stripe virus (RSV), is one of the most serious rice diseases in temperate and subtropical regions of the world. Since RSV is not transmissible mechanically, an insect transmission test was the original basis for identification of the viral population and cultivar resistance. A simple, rapid and reliable method is described by which virus-free small brown plan...
