چکیده
Abstract Excessive inflammatory responses induced upon SARS-CoV-2 infection are associated with severe symptoms of COVID-19. Inflammasomes activated in response to also COVID-19 severity. Here, we show a distinct mechanism by which N protein promotes NLRP3 inflammasome activation induce hyperinflammation. facilitates maturation proinflammatory cytokines and induces cultured cells mice. Mechanistically, interacts directly protein, the binding ASC, assembly. More importantly, aggravates lung injury, accelerates death sepsis acute inflammation mouse models, IL-1? IL-6 Notably, N-induced injury cytokine production blocked MCC950 (a specific inhibitor NLRP3) Ac-YVAD-cmk (an caspase-1). Therefore, this study reveals excessive responses.

Genipin suppresses NLRP3 inflammasome activation through uncoupling protein-2.
Incomplete clearance of apoptotic cells and reactive oxygen species (ROS) release are known to trigger inflammasome activation causing severe inflammation in acute lung injury and various metabolic and autoimmune diseases. Moreover, it has been reported that apoptotic cell clearance and ROS-mediated apoptosis critically depend on mitochondrial uncoupling protein-2 (UCP2). However, the relations...
متن کاملPKM2-dependent glycolysis promotes NLRP3 and AIM2 inflammasome activation
Sepsis, severe sepsis and septic shock are the main cause of mortality in non-cardiac intensive care units. Immunometabolism has been linked to sepsis; however, the precise mechanism by which metabolic reprogramming regulates the inflammatory response is unclear. Here we show that aerobic glycolysis contributes to sepsis by modulating inflammasome activation in macrophages. PKM2-mediated glycol...
متن کاملThe Adaptor MAVS Promotes NLRP3 Mitochondrial Localization and Inflammasome Activation
NLRP3 is a key component of the macromolecular signaling complex called the inflammasome that promotes caspase 1-dependent production of IL-1β. The adaptor ASC is necessary for NLRP3-dependent inflammasome function, but it is not known whether ASC is a sufficient partner and whether inflammasome formation occurs in the cytosol or in association with mitochondria is controversial. Here, we show ...
متن کاملWhole-inactivated SARS-CoV-2 vaccines: from fear to hope
Undoubtedly, vaccination can be one of the promising approaches to control infectious diseases such as the COVID-19 pandemic. Inactivated viral vaccines have a history of "vaccine-induced enhanced disease", which may occur when neutralizing antibodies bind to viral antigens without blocking or clearing the infection. This can cause additional inflammation through the mechanisms described for ot...
متن کاملUpregulated NLRP3 Inflammasome Activation in Patients With Type 2 Diabetes
Despite the recent attention focused on the roles of the nucleotide binding and oligomerization domain-like receptor family pyrin domain-containing 3 (NLRP3) inflammasome in the pathogenesis of type 2 diabetes, little is known about the ex vivo profile of inflammasome activation in type 2 diabetic patients. In this study, we investigated patterns of NLRP3 inflammasome activation in monocyte-der...
