RP-HPLC-Based Bioanalytical Approach for Simultaneous Quantitation of Cinnarizine and Domperidone in Rat Plasma

An accurate, precise and sensitive reverse-phase high-performance liquid chromatography (RP-HPLC) bioanalytical approach was developed for the simultaneous estimation of cinnarizine (CIN) domperidone (DOM) in rat plasma using irbesartan (IRB) as an internal standard (IS). The proposed RP-HPLC validated per latest ICH M10 guidelines. analytes (CIN DOM) IS were extracted from samples protein precipitation strategy. Chromatographic separation is accomplished by a C18 SunfireTM (5 µm, 250 mm × 4.6 mm) analytical column, isocratic mobile phase consisting acetonitrile-methanol 30:70 proportions at flow rate 1 mL/min. detection all three constituents recorded wavelength 270 nm with UV detector. DOM, CIN eluted 3.2, 4.5 6.1 min, respectively, utilizing total run time 10 min. lower limit quantification (LLOQ) 5 ng/mL DOM plasma. linear 5–200 range DOM. recovery method greater than 95%, relative uncertainty less 2%, indicating that accurate precise. established 1.1 1.7 created found to be robust passed validation criteria; thus, can employed successfully assessment
