Rotavirus Samples processing using Indigenously Developed Molecular Transport Medium (IMTM)

Objective: To perform comparative diagnostic study with rotavirus infected stool samples for efficacy ofindigenously prepared molecular transport medium (IMTM).Study Design: It was an experimental study.Place and Duration of Study: The conducted at Department Microbiology University Karachi,Karachi Pakistan from June to July 2021.Materials Methods: current based on hospital surveillance rotavirusgastroenteritis among children <5 years age, were collected in commercially available viraltransport medium. Samples known positive /negative through PREMIER Rota clone Enzyme-Linked Immunosorbent Assay (ELISA) kit test. Total 64 selected evaluate theefficacy indigenous (IMTM). Stool spiked IMTM andcommercial viral (CVTM). RNA extraction Real-time PCR performed byQIAmp house assay respectively as per manufacturer' instructions. Similar method extractionand Real-Time both type adopted.Results: Comparative analysis Cycle threshold (Ct) values paired (IMTM vs CVTM) performed.Real-Time (RT-PCR) negative results consistent each pair (p<0.0001).Conclusion: indigenously formulated found capable sample collection, transportation andprocessing detection PCR.
