Returning to Nature for the Next Generation of Antimicrobial Therapeutics

چکیده
Antibiotics found in and inspired by nature are life-saving cures for bacterial infections have enabled modern medicine. However, the rise resistance necessitates discovery development of novel antibiotics alternative treatment strategies to prevent return a pre-antibiotic era. Once again, can serve as source new therapies form natural product microbiota-based therapies. Screening soil bacteria, particularly actinomycetes, identified most used clinic today, but rediscovery existing molecules prompted shift away from discovery. Next-generation sequencing technologies bioinformatics advances revealed untapped metabolic potential harbored within genomes environmental microbes. In this review, we first highlight current mining chemical space, including approaches activate silent biosynthetic gene clusters situ culturing methods. Next, describe how using live microbes simultaneously leverage many diverse antimicrobial mechanisms treat disease impressive efficacy fecal microbiome transplantation consortia on infection. Nature-provided some important drugs human history, show that will continue offer valuable inspiration next generation antibacterial therapeutics.

Next-Generation Therapeutics for IBD
Various novel drugs have recently been evaluated in clinical trials showing promising effects in patients with inflammatory bowel disease (IBD). Here, we summarize the recent literature in the area of emerging therapies in the field of IBD, with specific focus on anti-integrin antibodies, such as vedolizumab (anti-α4β7) and etrolizumab (anti-rhuMAb β7), and the Janus kinase (JAK) inhibitor tofa...
متن کاملNext generation therapeutics for Alzheimer's disease
To date, no truly effective therapy has been developed for Alzheimer's disease or mild cognitive impairment. In searching for new approaches that may succeed where previous ones have failed, it may be instructive to consider the successful therapeutic developments for other chronic illnesses such as cancer and human immunodeficiency virus.
متن کاملBringing next-generation therapeutics to the clinic through synthetic biology.
Recent advances in synthetic biology have created genetic tools with the potential to enhance the specificity, dynamic control, efficacy, and safety of medical treatments. Interfacing these genetic devices with human patients may thus bring about more efficient treatments or entirely new solutions to presently intractable maladies. Here we review engineered circuits with clinical potential and ...
متن کاملAntimicrobial Peptidomimetics: Reinterpreting Nature to Deliver Innovative Therapeutics
PALETTE OF EXTRAORDINARY COLORS Virtually all multicellular organisms must ward off pathogenic microbes in order to survive and thrive on this planet. To accomplish this, most metazoans rely on geneencoded antimicrobial peptides (AMPs) as an essential part of their innate immune system. The role played by AMPs – and by the other umoral and cellular components of innate immunity – is particularl...
متن کاملNext-generation antimicrobial susceptibility testing.
Antimicrobial resistance has emerged as one of the most-significant health care problems of the new millennium, and the clinical microbiology laboratory plays a central role in optimizing the therapeutic management of patients with infection. This minireview explores the potential value of innovative methods for antimicrobial susceptibility testing of microorganisms that could provide valuable ...
