The effect of resveratrol on the sphingolipid metabolism has been investigated in ras-transformed 3T3 fibroblasts. Resveratrol is a phytoalexin great medico-biological significance, which reported to exhibit significant beneficial effects such as antioxidant, lipid-lowering, anti-neoplastic, etc. Sphingolipids are functionally active lipid molecules, implicated important cellular processes, proliferation, migration, apoptosis and transmembrane signalling, among others. Although there many investigations devoted influence various types cancer cells, mechanism this pathway remains unclear. present studies showed that major sphingolipid, sphingomyelin, was decreased membranes from resveratrol-treated fibroblasts, whereas ceramide, pro-apoptotic factor, elevated due neutral sphingomyelinase up-regulation. In addition, level sphingosine, product ceramidase, increased content sphingosine-1-phosphate, related cell lowered treatment, an finding especially for cells. balancing enzyme pathway, sphingosine kinase, produces down-regulated oncogene-transformed conclusion, results clearly show treatment fibroblasts with induces changes representatives metabolic shifting “sphingolipid rheostat” towards prevalence proapoptotic factor ceramide. observations can be applied development complex anti-tumour therapeutic strategies.
