Repercussions of high extracellular K +on T-cell antitumor responses

چکیده

Abstract Tumours create an immunosuppressive microenvironment that limits the antitumor function of infiltrating T-cells. Dying/necrotic tumor cells extrude intracellular potassium (K +) into local extracellular milieu, raising K +([K +] e) by 8–15 fold, which then accumulates in tumor-infiltrating Here, we determined functional impact high-[K e(50 mM) on human Using molecular assays, demonstrated prolonged exposure to ereduced CD3ɛ and CD25 (intermediate activation marker) surface expression, suggesting weakened T-cell activation. High-[K eperturbed receptor signalling downstream glycolysis pathway. metabolomics analysis, identified dysregulation pathways involving fatty acid oxidation, TCA cycle choline metabolism under e. Exposure eimpeded motility skewed polarization towards T-helper 2 (Th2) regulatory (T reg) subsets. A similar Th2 subset trend, although not significant, was seen core patient-derived biopsies (n=7) along with amassment T regcells intratumorally. Collectively, our findings substantiate effects eon functions, underlines potential use +efflux channel activators reversing dampened responses. HPV16 tumor associated macrophages suppress antitumor T cell responses.

PURPOSE High-risk human papillomavirus (HPV) is the main etiologic factor for cervical cancer. The severity of HPV-associated cervical lesions has been correlated to the number of infiltrating macrophages. The objective of this work is to characterize the role of tumor-associated macrophages (TAM) on the immune cellular response against the tumor. EXPERIMENTAL DESIGN We used the HPV16 E6- and...

Effect of LIGHT Adjuvant on Kinetics of T-Cell Responses Induced by HSV-1 DNA Immunization

Background: Studies on efficacy of various vaccines that prevent or reduce the primary and recurrent HSV-1 infection have demonstrated the importance of cellular immunity for protection against the infection. We previously used DNA vaccination to induce cellular immunity against HSV-1 infection in mice. Objective: The aim of our study was to evaluate the effect of LIGHT, a member of TNF super f...

CD25+ T cell depletion impairs murine squamous cell carcinoma development via modulation of antitumor immune responses.

Squamous cell carcinoma (SCC) constitutes a microenvironment that could modulate the antitumor immune response. Also, tumor-infiltrating lymphocytes are believed to play complex regulatory roles in antitumor immunity against SCC. The presence of regulatory T cells (Tregs) has been associated with the suppression of tumor-reactive T cells. However, the underlying mechanism for this T cell dysfun...

LSECtin expressed on melanoma cells promotes tumor progression by inhibiting antitumor T-cell responses.

Therapeutic antibodies that target T-cell co-inhibitory molecules display potent antitumor effects in multiple types of cancer. LSECtin is a cell surface lectin of the DC-SIGN family expressed in dendritic cells that inhibits T-cell responses. LSECtin limits T-cell activity in infectious disease, but it has not been studied in cancer. Here we report the finding that LSECtin is expressed commonl...

Role of the complement system in NK cell-mediated antitumor T-cell responses

The role of the complement system in oncogenesis and tumor progression remains poorly understood. We have recently demonstrated that the induction of a tumor-specific CD8+ T-cell response is improved upon transient inhibition of the complement system, which is coupled to an increased availability of natural killer cells. The complement system may therefore turn out to constitute a promising tar...
