Relationships between DNA repair and RTK-mediated signaling pathways

چکیده
Receptor Tyrosine Kinases (RTK) are an important family involved in numerous signaling pathways essential for proliferation, cell survival, transcription or cell-cycle regulation. Their role and involvement cancer survival have been widely described the literature, generally associated with overexpression and/or excessive activity pathology. Because of these characteristics, RTKs relevant targets fight against cancer. In last decade, increasingly works describe RTK modulation DNA repair, thus providing evidence relationship between protein actors repair pathways. this review, we propose a summary as potential modulators double-stranded order to put forward new lines research aimed at implementation therapeutic strategies targeting both RTK-mediated

Targeting RTK Signaling Pathways in Cancer
The RAS/MAP kinase and the RAS/PI3K/AKT pathways play a key role in the regulation of proliferation, differentiation and survival. The induction of these pathways depends on Receptor Tyrosine Kinases (RTKs) that are activated upon ligand binding. In cancer, constitutive and aberrant activations of components of those pathways result in increased proliferation, survival and metastasis. For insta...
متن کاملLapatinib, a preventive/therapeutic agent against mammary cancer, suppresses RTK-mediated signaling through multiple signaling pathways.
Activation of receptor tyrosine kinases (RTK) plays a key role in the prognosis of mammary cancer. Lapatinib is a small molecule dual RTK inhibitor that targets epidermal growth factor receptor (EGFR) and human epidermal growth factor receptor 2 (HER2). Identifying the protein targets involved in the effects of lapatinib and other RTK inhibitors might help determine why preventive efficacy vari...
متن کاملUntangling the relationships between DNA repair pathways by silencing more than 20 DNA repair genes in human stable clones
Much effort has long been devoted to unraveling the coordinated cellular response to genotoxic insults. In view of the difficulty of obtaining human biological samples of homogeneous origin, I have established a set of stable human clones where one DNA repair gene has been stably silenced by means of RNA interference. I used pEBVsiRNA plasmids that greatly enhance long-term gene silencing in hu...
متن کاملCanonical RTK-Ras-ERK signaling and related alternative pathways
Receptor Tyrosine Kinase (RTK)-Ras-Extracellular signal-regulated kinase (ERK) signaling pathways control many aspects of C. elegans development and behavior. Studies in C. elegans helped elucidate the basic framework of the RTK-Ras-ERK pathway and continue to provide insights into its complex regulation, its biological roles, how it elicits cell-type appropriate responses, and how it interacts...
متن کاملMultiple RTK pathways downregulate Groucho-mediated repression in Drosophila embryogenesis.
RTK pathways establish cell fates in a wide range of developmental processes. However, how the pathway effector MAPK coordinately regulates the expression of multiple target genes is not fully understood. We have previously shown that the EGFR RTK pathway causes phosphorylation and downregulation of Groucho, a global co-repressor that is widely used by many developmentally important repressors ...
