چکیده
Skeletal muscle differentiation is a tightly regulated process, and the importance of mammalian SWI/SNF (mSWI/SNF) chromatin remodeling family for regulation genes involved in skeletal myogenesis well-established. Our prior work showed that bromodomains mSWI/SNF ATPases BRG1 BRM contribute to by facilitating binding enzymes regulatory regions myogenic other target genes. Here, we report pathway analyses differentially expressed from study identified an additional role via Wnt signaling pathway. The has been previously shown be important development. To investigate pathway, individual dual knockdowns were performed followed RNA-sequencing. results show BRG1, but not BRM, regulator components downstream Reactivation stabilization β-catenin could rescue defect gene expression due knockdown or bromodomain inhibition using specific small molecule inhibitor, PFI-3. These demonstrate required upstream function. Chromatin immunoprecipitation at promoters component β-catenin, which provides further mechanistic insight transcriptional these

Interaction of viral oncogenic proteins with the Wnt signaling pathway
It is estimated that up to 20% of all types of human cancers worldwide are attributed to viruses. The genome of oncogenic viruses carries genes that have protein products that act as oncoproteins in cell proliferation and transformation. The modulation of cell cycle control mechanisms, cellular regulatory and signaling pathways by oncogenic viruses, plays an important role in viral carcinogenes...
متن کاملThe Canonical Wnt Signaling (Wnt/β-Catenin Pathway): A Potential Target for Cancer Prevention and Therapy
Precise regulation of signal transduction pathways is crucial for normal animal development and for maintaining cellular and tissue homeostasis in adults. The Wnt/Frizzled-mediated signaling includes canonical and non-canonical signal transduction pathways. Upregulation or downregulation of the canonical Wnt-signaling (or the Wnt/β-Catenin signal transduction) leads to a variety of human diseas...
متن کاملA Wnt- and -catenin-dependent pathway for mammalian cardiac myogenesis
Acquisition of a cardiac fate by embryonic mesodermal cells is a fundamental step in heart formation. Heart development in frogs and avians requires positive signals from adjacent endoderm, including bone morphogenic proteins, and is antagonized by a second secreted signal, Wnt proteins, from neural tube. By contrast, mechanisms of mesodermal commitment to create heart muscle in mammals are lar...
متن کاملThe Role of Mammalian Target of Rapamycine Signaling Pathway in Central Nervous System Cancers: A Review
Mammalian mechanistic target of rapamycine (mTOR) is a conserved serine/threonine kinase in the cellular PI3K/Akt/mTOR signaling pathway. This pathway is modified by cellular alterations such as level of energy, growth factors, stresses, as well as the increased environmental level of cancerous cytokines. In general, increase of this kinase protein function is seen in various types of cancers, ...
متن کاملAngiopoietin-like protein 8 (betatrophin) may inhibit hepatocellular carcinoma through suppressing of the Wnt signaling pathway
Objective(s): Hepatocellular carcinoma (HCC) is one of the leading fatal neoplasms and the most common primary liver malignancy worldwide. Peptide hormone ANGPTL8 (betatrophin) may act as an important regulator in HCC development through the Wnt/β-catenin pathway. We aimed to evaluate the effects of recombinant ANGPTL8 on Wnt/β-catenin signaling in human liver carcinom...
