Regulation of bta-miRNA29d-3p on Lipid Accumulation via GPAM in Bovine Mammary Epithelial Cells

MicroRNAs (miRNAs) are small RNA molecules consisting of approximately 22 nucleotides that engaged in the regulation various bio-processes. There is growing evidence miR-29 a key regulator hepatic lipid metabolism. Mimics and inhibitors bta-miRNA29d-3p were transiently transfected bovine mammary epithelial cells (BMECs) to reveal on accumulation BMECs. Results showed overexpression significantly inhibited expression genes related triglyceride (TAG) synthesis, namely DGAT1 mitochondrial glycerol-3-phosphate acyltransferase (GPAM, p < 0.01) down-regulated TAG levels (p 0.05). The fatty acid synthesis desaturation-related FASN, SCD1, ACACA, transcription factor SREBF1 also decreased. Interference increased GPAM, DGAT1, 0.01), upregulated concentration cells. Furthermore, luciferase reporter assay confirmed GPAM direct target bta-miRNA29d-3p. In summary, modulates metabolism by regulating BMECs targeting GPAM.
