Recurrent events modelling of haemophilia bleeding events

چکیده
Abstract A pharmacokinetic–pharmacodynamic (PK-PD) approach is developed for modelling the recurrent bleeding events in patients with severe haemophilia to investigate relationship between factor VIII plasma activity level and instantaneous risk of a bleed. The model incorporates patient-level pharmacokinetic (PK) information obtained through measurements taken prior study which are used fit non-linear mixed-effects two-compartment PK model. Dosing times within combined provide estimated all patients, as time-dependent covariate Methods correct attenuation effects that would otherwise arise due discrepancy true VIII. In contrast existing methods proposed such data, count data regression or time-to-event analysis, new method allows be current performance estimators assessed via simulation found outperform naive estimator, treats levels if they were measured without error, both terms bias mean squared error.

Dynamic Frailty and Change Point Models for Recurrent Events Data
Abstract. We present a Bayesian analysis for recurrent events data using a nonhomogeneous mixed Poisson point process with a dynamic subject-specific frailty function and a dynamic baseline intensity func- tion. The dynamic subject-specific frailty employs a dynamic piecewise constant function with a known pre-specified grid and the baseline in- tensity uses an unknown grid for the piecewise ...
متن کاملModelling recurrent events: a tutorial for analysis in epidemiology.
In many biomedical studies, the event of interest can occur more than once in a participant. These events are termed recurrent events. However, the majority of analyses focus only on time to the first event, ignoring the subsequent events. Several statistical models have been proposed for analysing multiple events. In this paper we explore and illustrate several modelling techniques for analysi...
متن کاملModelling Recurrent Events for Improving Online Change Detection
The task of online change point detection in sensor data streams is often complicated due to presence of noise that can be mistaken for real changes and therefore affecting performance of change detectors. Most of the existing change detection methods assume that changes are independent from each other and occur at random in time. In this paper we study how performance of detectors can be impro...
متن کاملStatistical modelling for recurrent events: an application to sports injuries
BACKGROUND Injuries are often recurrent, with subsequent injuries influenced by previous occurrences and hence correlation between events needs to be taken into account when analysing such data. OBJECTIVE This paper compares five different survival models (Cox proportional hazards (CoxPH) model and the following generalisations to recurrent event data: Andersen-Gill (A-G), frailty, Wei-Lin-We...
متن کاملThromboembolic events, recurrent bleeding and mortality after resuming anticoagulant following gastrointestinal bleeding. A meta-analysis.
Gastrointestinal (GI) bleeding commonly complicates anticoagulant therapy. We aimed to systematically review the published literature to determine the risk of thromboembolism, recurrent GI bleeding and mortality for patients on long-term anticoagulation who experience GI bleeding based on whether anticoagulation therapy was resumed. We performed a systematic review of phase III randomised contr...
