Reconstruction of the Diaminopimelic Acid Pathway to Promote L-lysine Production in Corynebacterium glutamicum

The dehydrogenase pathway and the succinylase are involved in synthesis of L-lysine Corynebacterium glutamicum. Despite low contribution rate to production, is favorable for its simple steps potential increase production L-lysine. effect ammonium (NH4+) concentration on biosynthesis was investigated, results indicated that can be promoted a high NH4+ environment. In order reduce requirement NH4+, nitrogen source regulatory protein AmtR knocked out, resulting an 8.5% (i.e., 52.3 ± 4.31 g/L). Subsequently, upregulated by blocking or weakening tetrahydrodipicolinate (DapD)-coding gene dapD overexpressing ddh further enhance biosynthesis. final strain XQ-5-W4 could produce 189 8.7 g/L with maximum specific (qLys,max.) 0.35 0.05 g/(g·h) 5-L jar fermenter. titer qLys,max achieved this study about 25.2% 59.1% higher than original without enhancement pathway, respectively. serve as breakthrough point reconstruct diaminopimelic acid (DAP) promote production.
