Recombinant production of Thermus aquaticus single-strand binding protein for usage as PCR enhancer

Single-stranded DNA-binding (SSB) proteins play an important role in DNA metabolism involving replication, recombination, and repair all living beings. In molecular biology, SSB are used as enhancers to increase the efficiency specificity of PCR. Thermostable protein eliminates secondary structure or dimer formation significantly effectiveness amplification fragments. this study, it was ensured that gene thermophilic bacteria Thermus aquaticus (T. aquaticus) cloned into pET28b vector expressed E. coli BL21 (DE3) PLysE cells. Then, purification produced cells performed. 20 mg obtained from 1L bacterial culture, its purity more than 90%. It shown by PCR experiment study could efficiency.
