Alzheimer’s disease (AD) is one of the most common progressive neurodegenerative diseases that represents about 60-80 % total cases dementia all over world. Beta secretase (BACE1) a membrane-anchored aspartic protease which cleaves peptide bond between Met671-Asp672 APP (amyloid precursor protein) where its activity rate limiting step producing amyloid β, primary neuropathological hallmark Alzheimer's (AD). Therefore, inhibitors this enzyme have potential to be promising and attractive target for intervention potent therapeutics treatment AD. Actually, there are many challenges facing development effective drug-like BACE1 including penetration BBB selectivity against other proteases. However, efforts been devoted by pharmaceutical industries academia small size, potent, selective biologically active inhibitors. Till now, none FDA approved but some exhibited clinical potential.
