Reactivity Tuning of Metal‐Free Artificial Photoenzymes through Binding Site Specific Bioconjugation

چکیده

The design and development of artificial metal-free photoenzymes aims to combine the selectivity enzymatic reactions with benefits modern synthetic photocatalysts. Removing need for rare earth metals allowing milder reaction conditions, leading a more sustainable catalytic system. Here, we present novel photoenzyme by integrating an organophotocatalytic moiety based on donor-acceptor into steroid carrier protein (SCP-2L). SCP-2L possesses hydrophobic tunnel facilitating substrate binding in aqueous media. photocatalyst was site-selectively bound three variants, possessing non-native cysteine residue strategically placed around protein. modified were shown be selective oxidation organic sulfides giving up 192 turnovers.

Site-Specific Reactivity of Nonenzymatic Lysine Acetylation

Protein acetylation of lysine ε-amino groups is abundant in cells, particularly within mitochondria. The contribution of enzyme-catalyzed and nonenzymatic acetylation in mitochondria remains unresolved. Here, we utilize a newly developed approach to measure site-specific, nonenzymatic acetylation rates for 90 sites in eight native purified proteins. Lysine reactivity (as second-order rate const...

Engineering of DNA binding proteins into site-specific cutters: reactivity of Trp repressor-1,10-phenanthroline chimeras.

Trp repressor (TrpR) can be converted into a site-specific nuclease by chemical modification of the cysteine mutants TrpR D46C or TrpR E49C with 5-iodoacetamido-1,10-phenanthroline (OP). In the presence of cupric ion and 3-mercaptopropionic acid, TrpR-regulated operators are cleaved. The properties of these semisynthetic scission reagents have been compared. The E49C construct cleaves efficient...

Salt Effect Accelerates Site-Selective Cysteine Bioconjugation

Highly efficient and selective chemical reactions are desired. For small molecule chemistry, the reaction rate can be varied by changing the concentration, temperature, and solvent used. In contrast for large biomolecules, the reaction rate is difficult to modify by adjusting these variables because stringent biocompatible reaction conditions are required. Here we show that adding salts can cha...

Selective Tuning: Feature Binding Through Selective Attention

We present a biologically plausible computational model for solving the visual binding problem. The binding problem appears due to the distributed nature of visual processing in the primate brain, and the gradual loss of spatial information along the processing hierarchy. The model relies on the reentrant connections so ubiquitous in the primate brain to recover spatial information, and thus al...

Tuning Promoter Strength through RNA Polymerase Binding Site Design in Escherichia coli

One of the paramount goals of synthetic biology is to have the ability to tune transcriptional networks to targeted levels of expression at will. As a step in that direction, we have constructed a set of 18 unique binding sites for E. coli RNA Polymerase (RNAP) δ⁷⁰ holoenzyme, designed using a model of sequence-dependent binding energy combined with a thermodynamic model of transcription to pro...
